银屑1号含药血清对银屑病中性粒细胞分泌NE、trappin-2的影响

Effect of Serum Containing Yinxie Yihao Prescription on Neutrophil Elastase and Trappin-2 Expression by Neutrophils of Psoriasis

目的:观察银屑1号含药血清对银屑病患者体外培养的中性粒细胞分泌NE、trappin-2的影响,探讨其治疗银屑病的机制。方法:分离培养18例寻常型银屑病患者外周血中的中性粒细胞,实验设置6组:4个浓度的中药含药血清(即原液中药组、1/2中药组、1/4中药组、1/8中药组)、雷公藤多苷阳性对照、生理盐水空白对照。采用ELISA法检测各组分泌的NE、trappin-2表达水平。结果:NE表达水平:与生理盐水组比较,原液中药组NE表达水平最低,生理盐水组表达水平最高,随着中药含药血清浓度的降低,NE的表达水平随之升高(P<0.05)。trappin-2表达水平:与生理盐水组比较,原液中药组trappin-2表达水平最低,生理盐水组最高,随着中药含药血清浓度的升高,trappin-2的表达水平逐渐降低。NE/trappin-2比值方面:生理盐水组比值显著高于其余5组。原液中药、1/2、1/4、1/8中药组显著低于阳性对照雷公藤组与对照组生理盐水组,并且NE/Trappin-2的比值伴随中药含药血清浓度的升高而降低(P<0.05)。结论:银屑1号治疗银屑病的机理可能是通过降低银屑病患者外周血中中性粒细胞NE、trappin-2的表达,使两者达到平衡,从而减轻炎症反应。

Objective:To investigate the effect of Yinxie Yihao Prescription(YXYHP)on secretion of NE and trappin-2 in peripheral blood neutrophils of patients with psoriasis vulgaris and explore its mechanism for the treatment of psoriasis.Methods:The peripheral blood neutrophils of 18 patients with psoriasis vulgaris were isolated and cultured.Four different concentrations of YXYHP were given and the control group was treated with triptolide and physiological saline.The levels of NE and Trappin-2 were detected by ELISA.Results:NE expression level:Compared with the saline group,the NE expression level was the lowest in the original liquid Chinese medicine group,and the highest expression level was in the saline group.With the decrease of the serum concentration of the Chinese herbal medicine,the expression level of NE increased accordingly(P<0.05).Trappin-2 expression level:Compared with the saline group,the expression level of trappin-2 in the original solution group was the lowest,and that of the saline group was the highest.With the increase of serum concentration of TCM-containing drugs,the expression of Trappin-2 was gradually decreased.NE/Trappin-2 ratio:The ratio of saline group was significantly higher than the other 5 groups.The amount of NE/Trappin-2 ratio was significantly lower in the traditional Chinese medicine group,1/2,1/4 and 1/8 Chinese medicine group than those in the control group and the saline group of the control group(P<0.05).Conclusion:Yinxie Yihao Prescriptioncan can reduce the concentrations of NE and trappin-2 secreted by peripheral blood neutrophils in patients with psoriasis vulgaris,making a dynamic balance between NE and its inhibitor trappin-2,and thus reducing inflammatory response and treating psoriasis.
