基于AMPK、p38MAPK及COX Ⅳ探讨脾主肌肉与重症肌无力的关系 被引量:8

摘要 重症肌无力是一种难治性的自身免疫性疾病,其发病机制尚未明确。该病属于中医'痿证'范畴,临床治法以健脾益气为主。AMPK和p38MAPK信号通路能调节肌肉PGC-1α的表达,COX Ⅳ是线粒体氧化磷酸化过程中的关键酶,三者均与线粒体能量代谢密切相关。研究表明多种神经肌肉病均存在线粒体能量代谢异常,故文章从线粒体能量代谢相关信号通路AMPK、p38MAPK及COX Ⅳ入手,进一步探讨脾主肌肉与重症肌无力的关系。 Myasthenia gravis(MG)is a refractory autoimmune disease which pathogenesis is not clear yet.This disease belongs to flaccidity syndrome in traditional Chinese medicine and strengthening spleen and invigorating Qi is the major clinical treatment of it.AMP-activated Protein Kinase(AMPK)and p38 mitogen-activated protein kinase(p38 MAPK)signaling pathways regulate the expression of PGC-1 alpha(peroxisome proliferator-activated receptorγcoactivator-1α)in muscle and cytochrome c oxidase subunitⅣ(COXⅣ)is the critical enzyme in the process of mitochondrial oxidative phosphorylation.These three signaling pathways are closely related to mitochondrial energy metabolism.Many studies showed that there were abnormal mitochondrial energy metabolism in many neuromuscular diseases.Therefore,we further explore the relationship between spleen dominant muscle and myasthenia gravis from mitochondrial energy metabolism related signaling pathways AMPK,p38 MAPK and COXⅣ.
