慢性不完全性睡眠剥夺大鼠对海马TNF-α mRNA、IFN-γ mRNA表达及脾脏NK细胞活性的影响 被引量:2

摘要 目的:探讨慢性疲劳大鼠睡眠剥夺后对血清细胞因子,海马TNF-α mRNA、IFN-γ mRNA表达及脾脏NK细胞活性的影响。方法:采用负重力竭游泳法制备CFS大鼠模型,用小平台水环境法剥夺睡眠12 h,将20只SD大鼠随机分为对照组和睡眠剥夺组,连续应激21 d后,Morris水迷宫测量大鼠学习记忆能力;采用ELISA检测血清TNF-α、IFN-γ、TGF-β的含量;Rael time-PCR检测海马TNF-α mRNA、IFN-γ mRNA的表达;LDH释放法检测脾脏NK细胞活性。结果:与对照组相比,睡眠剥夺组体质量在7 d、21 d明显降低(P <0. 01),14 d降低(P <0. 05);力竭时间7 d增长(P <0. 05),14 d减少(P> 0. 05),21 d明显增长趋势(P> 0. 05);Morris水迷空间搜索实验,总路程、平均速度提高(P <0. 05),经过平台次数降低(P <0. 05);血清IFN-γ含量和海马IFN-γ mRNA表达升高(P <0. 05);脾脏NK细胞活性降低(P <0. 05)。结论:CFS出现的认知功能下降、抑郁等神经精神症状很可能是睡眠质量差导致,长期睡眠不足使NK细胞活性降低,是机体对致病微生物的抵抗能力崩溃的主要原因。 Objective:To investigate the effects of sleep deprivation on expressions of serum cytokines,TNF-αmRNA and IFN-γmRNA in hippocampus and NK cell activity in spleen.Methods:CFS rat models were prepared by negative gravity exhaust swimming and deprived of sleep for 12 hours by small platform water environment method.Twenty SD rats were randomly divided into control group and sleep deprivation group.Morris water maze test was performed after continuous stress for 21 days.Rats’learning and memory ability;Serum TNF-α,IFN-γand TGF-βlevels were measured by ELISA.TNF-αmRNA and IFN-γmRNA expressions in hippocampus were detected by Real time-PCR.NK cell activity in spleen was detected by LDH release assay.Results:Compared with the control group,sleep deprivation group’s body weight decreased significantly at 7 d and 21 d(P<0.01),decreased at 14 d(P<0.05).The exhaustion time increased at 7 d(P<0.05)and decreased at 14 d(P>0.05).There was a significant growth trend at 21 d(P>0.05).Morris water space search experiment’s total distance and average speed increased(P<0.05)and platform times decreased(P<0.05).Serum IFN-γcontent and hippocampal IFN-γmRNA expression were elevated(P<0.05).The spleen NK cell activity was decreased(P<0.05).Conclusion:The cognitive impairment,depression and other neuropsychiatric symptoms in CFS are likely to be caused by poor quality of sleep.Long-term sleep deprivation results in decreased NK cell activity,which is the main reason for the collapse of the body’s resistance to pathogenic microorganisms.
