山羊MSTN基因5′侧翼区序列克隆与分析 被引量:2

Cloning and Sequence Analysis about the 5′ Flanking Region of MSTN Gene in Goat

摘要 本文为了研究山羊MSTN基因对山羊生长和肌肉发育的作用以及在不同山羊品种间的遗传变异,先期对其5′侧翼区进行了克隆,并利用序列分析方法进行了分析。分析结果表明该克隆序列长度1344bp,包括5′调控区和5′UTR和部分编码区,含增强子序列、CAAT盒、TATA盒和起始密码子序列,并存在Oct-1、C/EBP和Nkx-2.5等多个转录因子的潜在结合位点;同时山羊与绵羊、奶牛、猪和小鼠的同源性分别为98.2%、96.1%、81.2%和59.4%。 In order to study the effect of MSTN gene on the grow th and development of muscle and genetic variation among breeds in goats, the 5′-flanking region of MSTN gene was cloned during the preliminary works and analyzed by DNA sequence analysis method in this paper .The results showed that the cloning DNA sequence was composed of 1344bp .According to sequence analysis ,it contains 5′-promoter, 5′-UTR and partial cds .Furthermore some regulatory elements ,such as enhancer sequence, CCAAT box , TATA boxes ,start codon sequence and some potential transcription factor binding sites ,for example Oct-1、C/EBP and Nkx -2.5, were found in the cloning sequence. The homologies of the sequence of goat with sheep, dairy cattle, porcine, and mice were 98.2%, 96. 1%, 81. 2% and 59.4%, respectively.
