摘要
目的:探讨姜黄素对代谢综合征模型大鼠脂肪细胞因子及p38丝裂原活化蛋白激酶(MAPK)表达的影响。方法:选取健康雄性SD大鼠112只,随机抽取15只为对照组,另外97只进行模型复制。造模组采用高糖高脂高盐饲料建立代谢综合征模型,对照组大鼠喂以普通标准饲料。模型建立后,饲料不变,模型组给予0.5%羧甲基纤维素钠溶液10 mL/kg,日1次,灌胃;二甲双胍组给予7 mg/mL二甲双胍混悬液10 mL/kg,日1次,灌胃;姜黄素低、中、高剂量组分别给予10、15、20 mg/mL姜黄素混悬液10 mL/kg,日1次,灌胃;对照组给予生理盐水10 mL/kg,日1次,灌胃。各组大鼠均给药8周。检测比较各组体质量、Lee’s指数、收缩压(SBP)、血脂、空腹血糖(FPG)、空腹胰岛素(FIns)、胰岛素抵抗指数(HOMA-IR)、脂肪细胞因子水平、脂肪组织p38 MAPK蛋白表达水平。结果:与模型组比较,各组大鼠体质量、Lee’s指数、SBP、血清甘油三酯(TG)、低密度脂蛋白(LDL-C)、FPG、FIns、HOMA-IR及瘦素(LP)、内脂素(VF)较低,棕色脂肪组织、皮下脂肪组织及肾周脂肪垫p38 MAPK蛋白表达水平较低,且姜黄素低剂量组>姜黄素中剂量组>姜黄素高剂量组、二甲双胍组,差异均有统计学意义(P<0.05)。与模型组比较,各组大鼠血清高密度脂蛋白(HDL-C)、脂联素(ADP)较高,且姜黄素低剂量组<姜黄素中剂量组<姜黄素高剂量组、二甲双胍组,差异有统计学意义(P<0.05)。结论:姜黄素能剂量依赖性减轻代谢综合征模型大鼠肥胖程度,降低血压、血脂、血糖水平,改善胰岛素抵抗状态,其作用可能与调节脂肪细胞因子分泌以及下调脂肪组织p38 MAPK蛋白表达有关。
Objective:To analysis the effects of curcumin on adipocytokines and p38 mitogen activated protein kinase(MAPK)expression in rats with metabolic syndrome.Methods:112 healthy male SD rats were selected,and 15 rats were randomly selected to be as the control group,and others were builded model.The model building group was established metabolic syndrome model by high glucose,high-fat and high salt diet,rats in the control group received common standard feed.After the model was established,the feed was unchanged,the model group was given 0.5%carboxymethyl cellulose solution 10 mL/kg,qd,gavage;metformin group was given 7 mg/mL metformin suspension 10 mL/kg,qd,gavage;curcumin low-dose group,middle-dose group,high-dose group were given 10 mg/mL,15 mg/mL,20 mg/mL curcumin suspension 10 mL/kg,qd,gavage;control group was given normal saline 10 mL/kg,qd,gavage.All rats in each group were treated for 8 weeks.The body mass,Lee’s index,systolic blood pressure(SBP),blood lipid,fasting plasma glucose(FPG),fasting insulin(FIns),insulin resistance index(HOMA-IR),and adipocytokine levels,expression of p38 MAPK protein in adipose tissue were detected and compared.Results:Compared with model group,the body mass,Lee’s indexes,SBP,serum triglyceride(TG),low density lipoprotein(LDL-C),FPG,FIns,HOMA-IR and leptin(LP)and laciotin(VF)were lower,the expression level of p38 MAPK protein in brown adipose tissue,subcutaneous adipose tissue and perirenal fat pad were lower in each group,and curcumin low-dose group>curcumin middle-dose group>curcumin high-dose group and metformin group,the differences were all statistically significant(P<0.05).Compared with the model control group,serum high density lipoprotein(HDL-C)and adiponectin(ADP)were higher in each group,and curcumin low-dose group<curcumin middle-dose group<curcumin high-dose group and metformin group,the differences were statistically significant(P<0.05).Conclusion:Curcumin can reduce the obesity degree of the metabolic syndrome model rats in a dose dependent manner,reduce blood pressure,blood lipid and blood sugar levels,improve the state of insulin resistance,the effect may be related to the regulation of adipocytokines secretion,and down regulating the expression of p38 MAPK protein in adipose tissue.
