乌日勒对兔眼视神经缺血再灌注损伤后视网膜中央动脉血液流变学和血流动力学影响

Effects of Wurile on Hemorheology and Hemodynamics of Central Retinal Artery after Optic Nerve Ischemia-reperfusion Injury in Rabbits

摘要 目的:探讨乌日勒对兔眼视神经缺血再灌注损伤后视网膜中央动脉血液流变学和血流动力学的影响,并观察乌日勒的保护作用。方法:将125只日本大耳白兔按照随机数字表法分为正常组、模型组、乌日勒组和原花青素组,采用输液瓶缓慢升高,使眼内形成50 mmHg的眼内压,并持续2 h的方法建立视网膜缺血再灌注模型。各组分别在造模1 h时测量视网膜中央动脉血液流变学和血流动力学参数,除正常组,其他各组于造模12、24、48、72 h后测定视网膜中丙二醛(MDA)含量与超氧化物歧化酶(SOD)活性。结果:与正常组比较,乌日勒组的全血黏度高切、全血黏度中切、全血黏度低切参数明显降低,而原花青素组全血黏度中切参数降低;除血浆黏度外,乌日勒组与模型组在其余血液流变学参数上比较差异有统计学意义(P<0.05),原花青素组全血黏度高切和全血黏度中切参数明显低于模型组,其余各血液流变学参数高于乌日勒组,比较差异有统计学意义(P<0.05)。与模型组比较,乌日勒组和原花青素组的舒张末期血流速度(EDV)明显升高,阻力指数(RI)明显降低;乌日勒组和原花青素组血流动力学参数上比较差异有统计学意义(P<0.05)。24、48、72 h时组内SOD活性明显高于12 h(P<0.05)。与模型组比较,乌日勒组及原花青素组在同时间点的MDA含量明显降低,SOD活性明显提高,差异有统计学意义(P<0.05)。结论:乌日勒可促进视网膜中央动脉血液流变学和血流动力学参数的改善,防治视网膜缺血再灌注损伤。 Objective:To investigate the effects of Wurile on hemorheology and hemodynamics of central retinal artery after optic nerve ischemia-reperfusion injury in rabbits,and to observe the protective effect of Wurile.Methods:125 Japanese white rabbits were divided into normal group,model group,Wurile group and procyanidins group according to random number table.The infusion bottle was slowly raised to form an intraocular pressure of 50 mmHg in the eye,and a retinal ischemia-reperfusion model was established by the method of 2 h.The rheology and hemodynamic parameters of the central retinal artery were measured at 1 h after each model.Except the normal group,the other groups measured the content of malondialdehyde(MDA)and superoxide dismutase(SOD)in the retina after 12,24,48 and 72 hours.Results:Compared with the normal group,the high blood viscosity,middle blood viscosity and low blood viscosity of Wurile group were significantly lower,while the proanthocyanidin group had a lower middle blood viscosity;in addition to the plasma viscosity,there was a significant difference in the remaining hemorheological parameters between the Wurile group and the model group(P<0.05).The procyanidins group was significantly lower than the model group in other hemorheological parameters,and the other hemorheological parameters were higher than the Wurile group.The difference was statistically significant(P<0.05).Compared with the model group,the EDV of the Wurile group and the proanthocyanidin group were significantly higher,and the RI was significantly lower.There was a statistically significant difference in blood flow parameters between the Wurile group and the proanthocyanidin group(P<0.05).The SOD activity between the groups at 24,48 and 72 h was significantly higher than that at 12 h(P<0.05).Compared with the model group,the MDA content in the Wurile group and the proanthocyanidin group was significantly lower at the same time,and the SOD activity was significantly increased,the difference was statistically significant(P<0.05).Conclusion:Wurile can promote the improvement of hemorheology and hemodynamics parameters of central retinal artery and prevent retinal ischemiareperfusion injury.
