摘要
目的:优选养心苦荞茶的最佳制备工艺并建立其成品中芦丁的含量测定方法,为该保健茶的开发提供一定的基础依据。方法:以干膏得率与甘草酸铵的转移率为指标,在单因素试验基础上,采用正交试验考察加水量、提取时间、提取次数对养心苦荞茶中淡竹叶甘草提取工艺的影响,对苦荞吸附淡竹叶、甘草药液比例进行考察,确定养心苦荞茶的最佳制备工艺;并采用HPLC测定养心苦荞茶中芦丁的含量。结果:优选出的最佳提取工艺为加16倍量水,回流提取3次,每次提取1 h;干膏得率20.96%,甘草酸铵转移率68.43%;苦荞与淡竹叶甘草药液的吸附比例为1∶1,养心苦荞茶所含芦丁的平均含量为3.62 mg/g。结论:优化出的提取工艺稳定,简便可行,可为养心苦荞茶的制备与生产提供参考。
Objective:To optimize the preparation technology of Yangxin Buckwheat tea,and to grasp the determination method of Rutin in its finished products,so as to provide certain basis for the development of the health tea.Methods:On the basis of single factor test,the effect of water amount,extraction time and extraction times on the extraction process of Radix Glycyrrhizae in Yangxin Buckwheat tea was investigated on the basis of the dry paste yield and the transfer rate of ammonium glycyrrhizinate.The preparation of tartary buckwheat leaves and liquorice liquid on tartary buckwheat was investigated and the preparation of buckwheat tea was prepared.Methods of the content of Rutin in Yangxin buckwheat tea was determined by HPLC.Result:The optimum extraction process is 16 times the amount of water,reflux extraction 3 times,extracting 1 h each time,the yield of dry paste 20.96%,the transfer rate of ammonium glycyrrhizinate 68.43%,the adsorption ratio of tartary buckwheat and licorice solution is 1∶1,and the average content of Yangxin Buckwheat tea is 3.62 mg/g.Conclusion:The optimized extraction process is stable,simple and feasible for the extraction and production of Yangxin Buckwheat tea.
