脑皮质撞击法构建颅脑创伤大鼠模型中设定参数的比较

摘要 目的利用脑皮质撞击仪设置不同参数,以构建不同损伤程度的颅脑创伤(TBI)大鼠模型,为相关参数的标准化提供更多参考依据。方法 64只大鼠随机分为8组,即V1、V2、V3、D1、D2、D3、假手术(S)和对照(C)组,利用电子脑皮质打击仪构建不同损伤程度TBI模型。术后48 h行改良神经功能损伤评分、网屏实验评分、旷场实验评分,并测定脑组织含水量。结果 V、D组大鼠改良神经功能损伤评分与C组及S组比较均升高,差异均有统计学意义(P <0.05)。网屏实验评分方面,除V1组外,其余各组与C组或S组比较均升高,差异有统计学意义(P <0.05);但D1与D2组比较差异无统计学意义,D2与D3组比较差异无统计学意义(P> 0.05)。旷场实验评分中,水平得分方面,V3、D1、D2、D3组与C组或S组比较均降低,且V3组高于D2和D3组,差异均有统计学意义(P <0.05);垂直得分方面,各模型组与C组或S组比较均降低,且V1、V2、V3组均高于D3组,差异有统计学意义(P <0.05);大便粒数方面,各组间差异均无统计学意义(P> 0.05)。脑组织含水量方面,与C组或S组比较,V2、V3、D1、D2和D3组脑组织含水量均升高,差异有统计学意义(P <0.05);但V3与D1组比较差异无统计学意义,D1组与D2组比较差异无统计学意义(P> 0.05)。结论在固定打击最低点持续时间均为200 ms的情况下,以2 mm的打击深度,4 m/s的打击速度构建轻度TBI大鼠模型,以2 mm的打击深度,5 m/s的打击速度构建中度TBI大鼠模型,以4 mm的打击深度,5 m/s的打击速度构建重度TBI大鼠模型,具有较好的成功率和区分度,该参数更为适合构建不同损伤程度的TBI大鼠模型。 Objective To provide more reference for the standardization of related parameters by setting different parameters for the cerebral cortex impactor to establish a traumatic brain injury(TBI)rat model for different degrees of injury.Methods 64 rats were randomly divided into eight groups,including V1,V2,V3,D1,D2,D3,sham(S)and control(C)group.A TBI model for different degrees of injury was established.Fortyeight postoperative groups were given neurological impairment score,screen experiment score,open-field test score and brain water content.Results When compared with the C or S group,the neurological severity scores of V and D groups increased,and the differences were statistically significant(P<0.05).As for screen test scores,except for V1 group,the other groups underwent an increase when compared with the C or S group,and the differences were statistically significant(P<0.05),but there was no significant difference between D1 and D2 group,or between D2 and D3 group(P>0.05).In the open field experiment,the horizontal scores in V3,D1,D2 and D3 groups were lower than those in C or S group,the scores in V3 group were significantly higher than those in D2 and D3 groups(P<0.05).The vertical score in each model group was lower than that in the C or S group,and the vertical scores in V1,V2 and V3 groups were significantly higher than those in D3 group(P<0.05).There was no significant difference in the number of fecal granules between these groups(P>0.05).In terms of brain water content,when compared with C or S operation group,V2,V3,D1,D2 and D3 groups had significantly higher content(P<0.05),but there was no significant difference between V3 and D1 group,or between D1 and D2 group(P>0.05).Conclusion Under the condition that the duration of the fixed strike is 200 ms,a mild TBI rat model can be constructed with a striking depth of 2 mm and a striking speed of 4 m/s,a moderate TBI rat model can be constructed with a striking depth of 2 mm and a striking speed of 5 m/s,and a severe TBI rat model can be constructed with a striking depth of 4 mm and a striking speed of 5 m/s,which is more suitable for constructing different degrees of TBI rat model due to a better success rate and discrimination degree.
