摘要
目的 评价中药抗血吸虫病肝纤维化的效果。方法 用日本血吸虫尾蚴感染新西兰白兔 ,造成肝纤维化模型。 12周后服吡喹酮杀虫治疗 ,继以复方中药煎剂灌胃 ,并与秋水仙碱及生理盐水组 (对照组 )比较 ,观察血清前胶原 (PC )、 型胶原 ( - C)、透明质酸 (HA)、层粘连蛋白 (L N)的含量 ,并用天狼红染色及免疫组化法观察肝组织胶原蛋白的沉积、分布和含量 ;用原位杂交检测肝组织 型前胶原 m RNA的表达。结果 中药组、秋水仙碱组血清纤维指标较病理对照组显著下降 ,肝组织胶原蛋白含量和 型胶原 m RNA的表达也明显减少 ,但均未降至正常对照组水平。结论 补气。
Objective To observe the anti fibrosis effect of compound Chinese herbs in rabbits infected with Schistosomiasis japonicum. Methods The rabbits were infected with Schistosoma japonicum to induce rabbit model of hepatic fibrosis. The rabbits were treated with Praziquantel after 12 weeks and then were divided into 4 groups: the first group was treated with compound Chinese herbs, the second with Colchicine, the third with saline, and the fourth without infection also with saline. The PCⅢ, Ⅳ, HA, LN in serum were detected by RIA. The quantitative analysis of collagen in liver tissue was made by Picric Sirius red staining and immunohistochemical staining. The expression of procollagen Ⅰ mRNA was detected by in situ hybridization. Results The levels of PCⅢ, Ⅳ, HA, LN in the group treated with compound Chinese herbs and the group with Colchicine were significantly lower than those in the model control group with saline. The quantity of collagen and the expression of procollagen I mRNA were also significantly reduced. All above were still higher than those in normal control group. Conclusion The compound Chinese herbs have the anti fibrosis effect on the experimental model of schistosomiasis induced hepatic fibrosis in rabbits.
