人肺癌GSTM1基因多态性与p53突变的相关性 被引量:2

摘要 目的 探讨谷胱苷肽 S转移酶 μ1(GSTM1)基因的多态性与抑癌基因 p5 3突变在人肺癌发生中的关系。方法 收集 4 8例人肺癌标本 ,HE染色做病理诊断 ,抽提基因组 DNA,PCR扩增 GSTM1基因的第 4外显子 ,聚丙烯酰胺凝胶电泳分析基因的多态性。对相应的肺癌标本用免疫组织化学的方法做突变型 P5 3蛋白表达的检测 ,分析二者的相关性。结果 在人的肺癌组织中 ,GSTM1基因的缺失率较高 ,达 5 4 .2 % ;突变型 P5 3蛋白总的表达率为 5 6 .3% ,其中GSTM1基因缺失者与非缺失者的 p5 3突变率分别为 73.1%和 36 .4 % ,二者之间有显著性差异 ,P<0 .0 5。结论 在人肺癌组织中 ,GSTM1基因的多态性与 p5 3基因突变明显相关 ,GSTM1基因的缺失可能会导致 p5 Objective To study the relationship between the polymorphism of glutathione S transferase μ1 (GSTM1) and suppressor gene p53 mutation in human lung cancer. Methods A total of 48 human lung cancer specimens were collected and stored in liquid nitrogen. All specimens were subjected to the diagnosis of HE staining. DNA of human lung cancer specimens were extracted, and GSTM1 4 exon gene was amplified by PCR, the polymorphism of GSTM1 gene was analyzed by polyacrylamide gel eletrophoresis, and mutant P53 protein expression in the corresponding lung cancer specimens was detected by immunohistochemistry method. Results In human lung cancer specimens, GSTM1 gene deletion(0/0)was up to 54 2 % and the expressing rate of mutant P53 protein was 56 3 %. The P53 mutant rate was 73 1 % in group of GSTM1 (0/0) and 36 4 % in group of GSTM1(+/+) or GSTM1 (+/0) respectively. There was significant difference between group of GSTM1 (0/0) and group of GSTM1 (+/+) or GSTM1 (+/0), P <0 05. Conclusion Polymorphism of GSTM1 is obviously related to p53 gene mutation, and the deletion of GSTM1 may lead to p53 gene mutation in Chinese lung cancer specimens.
