慢性缺氧对大鼠肺动脉平滑肌细胞形态的影响

摘要 目的 探讨慢性缺氧引起缺氧性肺血管收缩反应降低的机制。方法 采用计算机成像及图像分析系统 ,观察慢性缺氧培养的大鼠肺内动脉平滑肌细胞形态学变化的情况。结果 慢性缺氧后肺动脉平滑肌细胞中缺氧敏感型细胞的比例明显减小 ,中间混合型细胞比例明显增高 ,而缺氧不敏感型细胞的比例不变 (P<0 .0 5 )。结论 慢性缺氧可直接使肺动脉平滑肌细胞表型转化 ,由缺氧敏感细胞向中间混合型细胞转化。这可能是慢性缺氧时缺氧性肺血管收缩反应降低的机制之一。 Objective To investigate the mechanism of chronic hypoxia induced decreased HPV. Methods By using image analysis system the morphological changes of porcine pulmonary artery smooth muscle cells (PASMC) cultured under sustained low O 2 concentration were observed. Results Under chronic hypoxia, the proportion of the hypoxia sensitive type PASMC was decreased, while the mixed type cells were increased apparently ( P <0 05). Conclusion Chronic hypoxia could cause the transformation of PASMC's phenotype from hypoxia sensitve type to mixed one, which might be responsible for the decreased HPV induced by chronic hypoxia.
