摘要
目的 探讨姜黄素对卵巢癌细胞株 A2 780的生长抑制作用及其机制。方法 10~ 5 0μm ol/ L姜黄素作用 6~2 4 h后 ,MTT比色法检测细胞生长活性 ,流式细胞仪测定细胞周期时相变化 ,末端 Td T酶标记技术和 DNA L adder检测细胞凋亡 ;透射电镜观察细胞超微结构变化。结果 姜黄素能显著抑制卵巢癌细胞株 A 2 780的体外生长 ,呈时间与剂量依赖性 ;细胞周期阻滞于 G0 / G1 期 ;部分细胞出现凋亡形态学改变 ,凝胶电泳可见“梯形”条带 ,凋亡率为 6 .4 1%~ 2 8.4 8% (P<0 .0 1)。结论 姜黄素通过抑制细胞增殖、诱导细胞凋亡等机制 。
Objective To study the growth inhibition effects of curcumin on human ovary cancer cell line A2780 and the mechanisms. Methods After being treated with 10-50 μmol/L curcumin for 6-24 h, the growth activities of cancer cells were studied by MTT colorimetry. Cell cycle phases were inspected by flow cytometery (FCM). The apoptosis was detected by TUNEL and DNA ladder methods. The morphological changes of cancer cells were observed under electronic microscopy. Results Curcumin could effectively inhibit the in vitro growth of human ovary cancer cell line A2780 in time and dose dependent manners. Cell cycles were arrested at G 0 /G 1 phase. Partial cancer cells presented characteristic morphological changes of apoptosis, with obvious trapeziform bands on electrophoresis. The rates of apoptotsis were 6 41 %~28 48 % ( P <0 01). Conclusion Curcumin could significantly inhibit the growth of ovary cancer cells through blocking cellular proliferation and inducing apoptosis.
