摘要
目的 :探讨与bcl- 2翻译起始部位碱基互补的反义bcl- 2寡脱氧核苷酸 (oligodeoxynucleotide,ODN)对BcaCD885颊癌细胞热敏感性的影响。方法 :以脂质体Lepofectin为载体 ,转染 2 0 μM反义bcl- 2ODN于BcaCD885细胞内 ,阻断内源性bcl- 2蛋白表达 ,再以 43℃ 40min加热后 ,以流式细胞术—抗体及DNA分析对bcl- 2蛋白、bax蛋白和细胞凋亡进行检测。结果 :反义bcl- 2ODN阻断BcaCD885细胞内源性bcl- 2蛋白表达后 ,bax/bcl- 2升高 ,细胞凋亡率明显提高。结论 :阻断BcaCD885细胞内源性bcl- 2蛋白表达能提高其热敏感性。
Objective:To study the change of thermosensitivity by antisense oligodeoxynucleotides(ODN) which complements to translation-initiation site of human bcl-2 mRNA in BcaCD885 cells.Methods:After we ransfected 20μM antisense bcl-2 oligodeoxynucleotids into BcaCD885 cells through Lepofectin vector, we applied hyperthermic treatment at 43℃ for 40 minutes throuh water bath.We studied the expression of bcl-2 protein ,bax protein and apoptotic rate through flow cytometry analysis.Results:In due to inhibition of the expression of bcl-2 protein, the rate of bax to bcl-2 was raised and the apoptotic rate was improved.Conclusions:Antisense bcl-2 oligodeoxynucleotids can promote thermosensitivity of BcaCD885 cells.
