摘要
目的:研究小鼠受精卵冷冻解冻后的发育效果。方法:在20℃室温下,用玻璃化冷冻液(含乙二醇,丙二醇和葡聚糖)对昆明小白鼠受精卵进行一步法和二步法玻璃化冷冻保存。结果:当乙二醇为20%(V/V),丙二醇为20%(V/V),葡聚糖为20%(W/V)时,一步法的2min平衡组(72%)和二步法的1min平衡组(75%)发育效果最好,明显优于其它各实验组(P<0.01),但与对照组(98%)相比仍有极显著差异(P<0.01)。将玻璃化冷冻效果较好组的胚胎移植后,得到较高的妊娠产仔率,与新鲜胚胎的移植效果比较无显著差异。结论:实验所用的玻璃化冷冻方法效果较好,解冻后胚胎的发育率较高。
Objective:To study the preservation and development of mouse zygotes after being frozen and thawed. Methods: The zygotes of Kunming mice were vitrified in the media containing ethylene glycol, propylene glycol and dextran with one-step and two-step methods under 20C. Results: Better results were obtained when the zygotes were vitrified in the medium containing 20% (V/V) ethylene glycol,20% (V/V) propylene glycol and 20% (W/V) dextran and the time of e-quilibration lasted 2 min with one-step method and 1 min with two-step method. The developmental rates were 72% and 75% .respectively. These results were significantly better than other experimental groups,although they were significantly different from the controls of fresh embryos. Higher birth rates were got after the embryos frozen-thawed were transferred to recipients,but there was no significant differences compared with the group of fresh embryo transfer. Conclusion:Better effect of vitrification is achieved in this experiment,and higher developmental rate of thawed embryos is got.
