摘要
目的 :研究Fas及FasL在实验性大鼠肝促癌过程中的表达及作用。方法 :复制大鼠肝癌启动 /促进模型 ,用免疫组化方法检测大鼠肝促癌过程中Fas及FasL蛋白表达情况。结果 :正常组大鼠肝细胞Fas呈弱阳性表达 ,经启动处理后的各组 (高、中、低剂量组和启动组 )Fas表达增强 ,高、中剂量组在给予促癌物PB后 1周~ 1 5周 ,Fas呈强阳性表达 ,在给予PB1 5周后 ,Fas表达明显呈下降趋势 ,其余各组Fas表达未见明显改变。正常组和促癌组未见FasL蛋白表达 ,其余各组可见Fas表达。高剂量组和中剂量组在加促癌物PB后 1 5周 ,FasL蛋白表达明显增多。结论
Objective:To explore the expression and effect of Fas and FasL in rat liver tumor promotion by phenobarbital(PB) Methods:The rat liver initiating-promoting model was established by DEN treatment(initiating)followed by a selection phase(2-AFF treatment and partial hepatectomy)and then by PB-promoting Immunohistochemistry was used to investigate the expression of Fas and FasL Results:It was shown that Fas exfression level in all initiated groups(high-dose,middle-dose,low-dose and initiating group)was higher than that in normal control group;however,FasL was not observed in normal group or promoting group but expressed in other groups After 15 weeks of administration of PB,Fas protein in high-dose and middle-dose group declined significantly while FasL protein increased remarkably Conclusion:The results suggested that down-regulation of Fas and up-regulation of FasL might participate in the tumor promotion of rat liver by PB,while its mechanism still remain unclarified
