大鼠减体积肝移植模型的技术改进及肝再生的初步观察 被引量:12

An Improved Technique of Reduced-size Liver Transplantation in Rats and the Preliminary Assessment of Regeneration of the Graft

摘要 目的:建立稳定的大鼠减体积肝移植模型,并初步观察移植后肝脏再生的情况。方法:雄性SD大鼠,采用改良“二袖套法”建立大鼠减体积肝移植模型,无肝期(17.0±3.5)min。分别称取供、受体肝脏重量及供肝重量。术后1、4、7天处死大鼠,称取肝脏重量并取肝组织行组织学检查。结果:术后1周存活率85%。术后7天时受体肝脏重量已恢复至术前水平。组织学检查术后4、7天的肝组织可见二倍体及多倍体的肝细胞。结论:通过技术改进,提高了该模型建立的稳定性。大鼠减体积肝移植后供肝仍具有较强的再生能力。 Objective: To develop and reform a rat model of reduced-size liver transplantation and to investigate the liver regeneration after transplantation. Methods; The model was performed in SD rats. Two-cuff technique described by Kamada was used with some modification. The recipient underwent an anhepatic period of (17±3. 5)min. To weigh the liver weight of donor, recipient and graft respectively, recipient rats were killed at 1, 4 and 7 days after transplatation. The weight of graft was collected and the histopathologic examination of the liver was performed. Results: 85% of the grafted rats survived for 1 week. The grafted liver weight of recipient was similar to the preoperative level at 7 days after transplantation. On the 4th and 7th days after transplatation, the histopathologic examination of the grafted liver found diploid and polyploid hepatocyte. Conclusion: The modification of the techque provides a more practical and stable experimental model for the study of liver transplantation. The graft of the rats after reduced-size liver transplantation has a great ability of regeneration.
