胃癌细胞株SGC-7901及MGC-803中SURVIVIN基因转录与表达的鉴定

Identification of the expression of survivin gene in two human gastric adenocarcinoma cell lines(MGC80-3 and SGC-7901)

目的 探讨胃腺癌细胞株SGC 790 1及MGC 80 3中survivin基因转录与表达情况 ,为胃癌与survivin基因的相关研究提供帮助。方法 ①利用反转录聚合酶链反应 (RT PCR)及DNA序列测定技术检测两种细胞株中survivin基因mRNA转录情况。②采用免疫组织化学染色鉴定两种细胞株survivin基因有无蛋白表达及其表达程度。结果 从提取的胃癌细胞总RNA中扩增出 6 12bp的DNA片段 ,与预期的扩增片段大小相符 ;序列测定显示两细胞株的DNA扩增片段序列与基因库survivin基因mRNA序列完全一致 ;免疫组织化学染色显示两细胞株中均有survivin蛋白表达产物。结论 中国人胃腺癌细胞株SGC 790 1及MGC 80

Objective To investigate the expression of survivin gene in two human gastric adenocarcinoma cell lines (MGC 803 and SGC 7901). Methods Expression of survivin mRNA was evaluated by reverse transcriptase polymerase chain reaction (RT PCR) method and the amplified cDNA fragment was sequenced. Immunohistochemical assay was used to detect the expression of cellular survivin protein. Results The survivin mRNA transcription and high levels of survivin protein expression were detected in both of the two human gastric adenocarcinoma cell lines (MGC80 3 and SGC 7901).Conclusion The results indicated that the expression of survivin gene exists in the two cell lines MGC 803 and SGC 7901 and it may be useful for the further study of gastric cancer related to the survivin gene.
