摘要
目的 :探讨中药复方重肌灵免疫调节作用及其机理。方法 :建立Lewis大鼠EAMG模型。采用3H TdR法检测重肌灵对ConA、脂多糖 (LPS)诱导正常BALB C小鼠脾淋巴细胞增殖反应的影响 ,及重肌灵对ConA、N2 AchR诱导EAMG大鼠淋巴结细胞增殖反应的影响。ELISA法测定重肌灵对EAMG大鼠血清中N2 AchRab滴度及血清中IFN γ水平的影响。结果 :重肌灵对ConA、LPS诱导正常ALB C小鼠脾淋巴细胞增殖反应有明显的促进作用 (P <0 .0 5 ) ;对ConA诱导EAMG大鼠淋巴结细胞增殖反应有明显的促进作用 (P <0 .0 1 ) ,而对N2 AchR诱导EAMG大鼠淋巴结细胞增殖反应有明显抑制作用(P <0 .0 1 )。重肌灵对EAMG大鼠血清中N2 AchRab滴度及血清IFN γ水平有明显下调作用 (P <0 .0 1 )。结论 :重肌灵防治EAMG不是通过免疫抑制 ,而是利用它的免疫调节作用。机理可能是通过下调IFN γ水平 ,而抑制N2 AchR抗原诱导特异性免疫反应 。
AIM: To approach the immunoregulation function and mechanism o f compound zhongjiling. METHODS: EAMG models in Lews rats were e stablished. By using 3H-TdR method, the effect of zhongjiling to proliferation reaction of lymphocyte in normal BALB/C spleen of mice induced by ConA and Lps a nd the effect of compound zhongjiling to proliferation reaction of lymphocyte in lymph node of rats was examined; by using ELISA, the effect of zhongjiling to A chRab titre and the level of IFN-γ in serum of EAMG rats was examined. RESULTS: there was no significant improvement on the effect of zhongj iling to proliferation reaction of lymphocyte in normal BALB/C spleen of mice in duced by ConA and LPS(P< 0.05 ). The effect of compound zhongj iling on the proliferation reaction of lymphocyte showed a significant improveme nt (P< 0.01 ) in lymph node of EAMG rats induced by ConA, and the inhibition was induced by N 2AchR (P< 0.01 ). The AchRa b titre and the level of IFN-γ significantly reduced (P< 0.0 1 ) in serum of EAMG rats. CONCLUSIONS: The mechanism of compou nd zhongjiling is not through immunosuppression, but through restraining N 2Ach R antigen to induce specific immune reaction in prevention and treatment of EAMG .
