狼毒大戟对病毒性T细胞白血病的抑制作用 被引量:8

摘要 目的 研究中药狼毒大戟抗病毒性肿瘤机理。方法  6 15近交系小鼠 5 0只 ,随机分为5组 ,其中设肿瘤对照组 (Ⅰ ) ,2种剂量的治疗组 (Ⅱ、Ⅲ ) ,药物对照组 (Ⅳ )和正常对照组 (Ⅴ )。治疗组和肿瘤对照组皮下接种L6 15白血病肿瘤株 ,治疗组及药物对照组每天经胃分别给予狼毒水浸出液1.8g kg( 36mg)、3.0g kg( 6 0mg) ,连续 1周。检测外周血白细胞总数 ,观察肿瘤细胞凋亡形态特征及DNA电泳图谱 ,并检测肿瘤细胞c myc和ras基因的表达。结果  3.0g kg狼毒给药组外周血白细胞[( 0 .116 4± 0 .0 12 3)× 10 9 L]明显低于肿瘤对照组 [( 0 .2 199± 0 .10 99)× 10 9 L],P <0 .0 1。 1.8g kg和3 .0g kg狼毒药物组的细胞凋亡率 ( 2 3 .6 0 %± 2 .2 7% ) ,( 36 .40 %± 4.99% )均明显高于肿瘤对照组( 4.6 0 %± 0 .97% ) ,P <0 .0 1。在治疗组动物外周血中观察到大量的具有特征性的凋亡细胞和典型的DNA凋亡梯形电泳带。与肿瘤对照组相比 ,狼毒治疗组肿瘤细胞c myc和ras基因表达受到明显的抑制 (P <0 .0 1) ,表达强度也明显减弱。随着狼毒剂量增大 ,作用效果更为明显 (P <0 .0 1)。结论 狼毒大戟能抑制T淋巴细胞白血病的增殖 ,并可通过促进肿瘤细胞凋亡和抑制肿瘤细胞c myc和ras基因表达发挥抗病毒性肿瘤的作? Objective To investigate effects of Euphorbia fisheriana stead to cure T cell leukemia caused by retrovirus. Methods To divide 50 mice of 615 strain into 5 groups, leukemia group (Ⅰ), two treated groups (Ⅱ & Ⅲ), Euphorbia fisheriana stead control group without leukemia (Ⅳ), and normal group (Ⅴ). 615 mice from Ⅰto Ⅲ groups were inoculated by leukemia cells with retrovirus. Then Ⅱ, Ⅲ and Ⅳ group were given Euphorbia fisheriana stead water lixivium, the dosage was 1.8?g/kg, 3.0?g/kg, and 3.0?g/kg, respectively, orally once time a day for 1 week. The total number of WBC in peripheral blood was counted and apoptosis characteristics of tumor cells, such as apoptosis body and DNA electrophoresis were observed. c-myc and ras gene expressions were detected by histrochemical method. Results WBC number of treated group with 3.0?g/kg [(0.116?4± 0.012?3 )×109/L] was much lower than tumor group [(0.219?9±0.109?9)×109/L, ( P <0.01)]. The apoptosis rates in tumor cells of two treated groups [(23.60±2.27)%,(36.40±4.99)%] were higher than tumor group [(4.60±0.97)%] and P<0.01. The typical apoptosis morphology and DNA trapeziform stead were appeared in tumor cells of two treated groups. To compare with tumor group, the expressions of c-myc and ras in Euphorbia groups were less ( P <0.01).As the dosage increased, the effect of therapy was more remarkable(P< 0.01 ). Conclusion Euphorbia fisheriana stead can inhibit T leukemia cell proliferation and so is potential by promoting apoptosis and inhibiting c-myc/ras expression. [
