摘要
目的 探讨乳腺癌细胞凋亡与β1-整合素作用的关系。方法 人乳腺癌细胞SK -BR - 3在培养中以不同浓度的抗 β1-整合素单克隆抗体的作用下 ,观察其对TNF -α诱导的凋亡并计算凋亡率。结果 加入凋亡诱导剂后 ,凋亡细胞由正常生长的 2 85± 0 4 1增加到 2 5 6 2± 1 4 1(P <0 0 1) ;而加入抗 β1-整合素单克隆抗体的实验组凋亡细胞数明显减少 ,且随加入抗体浓度的增加而再度减少。结论 TNF -α对SK -BR - 3人乳腺癌细胞凋亡的诱导作用可能是由 β1-整合素参与的介导作用实现的 ,且存在着一种在一定范围内的量、效依赖关系。
Objective To study relationship between the apoptosis of breast carcinoma cells and the role of β 1-integrin. Methods The SK-BR-3 human breast carcinoma cells were incubated in 10% FBS1640 medium and anti-β 1-integrin mAb were treated with different concentration of these cells apoptosis induced by TNF-α and the apoptosis index counted. Results After apoptosis inductors were added apoptosis amount cells of increased from 2 85±0 41 to 25 62±1 41( P <0 01) in control group but in experiment groups the apoptosis cells with anti-β 1-integrin mAb decreased The higher the concentration,the lower the apaptosis cells will be. Conclusion The paper suggested the TNF-α induction effect on SK-BR-3 human breast carcinoma cell apoptosis maybe be caused by the β 1-integrin and having a dependent relationship on dosage. We support the report that integrins get involved in the cell apoptosis by series signal conductions.
