摘要 目的 探讨腺嘌呤致大鼠雄性不育的可能机制。方法 用含 0 5 %的腺嘌呤饲料饲喂大鼠 3 0d ,在实验第 3 0天活杀取睾丸组织 ,常规连续病理切片。观察睾丸组织的病理改变情况 ;免疫组化检测睾丸曲细精管TGF β1蛋白表达水平 ,脱氧核苷酸末端转移酶缺口末端标记法 (TUNEL)检测生精细胞凋亡。结果 实验组大部分曲细精管退行性变性 ,各级精细胞变性 ,数量减少 ;睾丸间质细胞和Sertoli细胞均可见TGF β1蛋白表达 ,而对照组只有睾丸间质细胞可见TGF β1蛋白表达 ,且实验组TGF β1蛋白表达强度较对照组为高 (P <0 .0 1) ;睾丸生精细胞凋亡细胞百分率较对照组明显增多 (P <0 .0 1)。结论 腺嘌呤诱导的大鼠雄性不育可能与TGF β1的抑制生精作用及促进生精细胞凋亡有关。 Objective To study the mechanism of adenine induced male infertility in rat.Methods Adenine induced male infertility was performed in rat by providing them with 0 5% adenine excessive diet.On days 30,the expressions of TGF β 1 in testis wered examined utilizing immunohistochemistry.the cell apoptosis of seminiferous tubules were examined by TUNEL method.Results The testis tissue was degeneration,the quantity of sperm was decrescent.TGF β 1 expression can been observed in interstitial cells and Sertoli cells,but the control group TGF β 1 expressions can been observed in interstitial cell only.Compared with the control group,the expressions of TGF β 1 in testis were increased( P <0.01),and the percentage of apoptosis cell in seminiferous tubules was increscented also( P <0.01).Conclusion The increscence of expression TGF β 1 and apoptosis cell in testis were related to the mechanism of adenine induced male infertility in rat.
