A Study on the Vitrificated Cryopreservation For Mouse Spleen Lymphocyte

试验研究了玻璃化冷冻法超低温保存小鼠脾淋巴细胞。结果表明 ,采用PVS2 玻璃化保存液 ,以二步法操作并以 3 7℃复苏后 ,经MTT法检测 ,细胞存活率可达 90 .8%以上。冻存后细胞能恢复生长并能形成杂交瘤细胞。

This experiment was to study the effects of vitrificated cryopreservation for mouse spleen lymphocyte. Frozen by two-step process with vitrificated solution PVS\-2 , and then thawed out in 37℃ , the survival rates for cells could attain 90.8% or above (based on the MTT assay) . The frozen cells regrew and could be formed lymphocytic hybridoma. Factors related to the survival rates of vitrification of mouse spleen lymphocyte are presented.
