三氧化二砷诱导表达带3蛋白膜段结构域K562细胞凋亡作用的研究(英文)

摘要 目的 探讨三氧化二砷诱导表达带 3蛋白膜段结构域K5 6 2细胞凋亡的作用。方法 将纯化的表达载体pYD1 mdb3转染K5 6 2细胞 ,WesternBlot检定有带 3蛋白膜段结构域的表达 ;通过光学显微镜、DNA电泳和线粒体膜电位的测定 ,观察三氧化二砷对表达带 3蛋白膜段结构域的K5 6 2细胞凋亡的影响。结果 表达带 3蛋白膜段结构域的K5 6 2细胞的上述凋亡特征均比未表达带 3蛋白的K5 6 2细胞明显。结论 表达带 3蛋白膜段结构域的K5 6 Objective To investigate the function of band 3 membrane domain during arsenic trioxide (As 2O 3) induced apoptosis of K562 cells.Methods The purified expression vector pYD1 mdb3 was transfected into K562 cells. The expressed band 3 membrane domain was identified by Western blot. The effect of As 2O 3 on K562 cells which expressed band 3 membrane domain was observed with light microscope, DNA electrophoresis, and mitochondrial membrane potential(MMP)assay. Results As 2O 3 induced more significant changes of apoptosis in K562 cells which expressed membrane domain of band 3 protein. Conclusion The K562 cells expressed band 3 membrane domain are more susceptible to As 2O 3.
