摘要
目的 探讨人脑胶质瘤中耐药蛋白P gp和MRP1表达情况。 方法 37例人脑胶质瘤标本 ,包括星形细胞瘤 2 5例 (纤维型 7例 ,原浆型 6例 ,间变型 12例 ) ,胶质母细胞瘤 12例 (多形性胶质母细胞瘤 4例 ) ;10例正常脑组织作为对照。P gp和MRP1的检测采用免疫组化ABC法。 结果P gp在肿瘤中阳性率为 2 4 3% ( 9/ 37) ,正常脑组织为 10 % ( 1/ 10 ) ,两者无统计学差异。MRP1在肿瘤中阳性率为 73% ( 2 7/ 37) ,正常脑组织无表达 ,两组间有明显差异。P gp和MRP1在标本血管内皮细胞中存在表达 ,其中P gp阳性率为 2 4 3% ( 9/ 37) ,MRP1为 35 1% ( 13/ 37)。 结论 人脑胶质瘤中 ,P gp和MRP1均有所表达 ,其中P gp较少 ,而MRP1明显增多 ,提示与耐药关系密切。在肿瘤血管内皮细胞中 ,两者存在异常 。
Objective To study the alteration of P glycoprotein (P gp) and multidrug resistance protein 1(MRP1)in gliomas.Methods Thirty seven gliomas were classified by WHO standard (1990) as astrocytomas (25 cases,including fibrillary,7 cases;protoplasmic,6 cases;anaplastic,12 cases),glioblastomas (12 cases,including GBM,4 cases).Ten normal brain tissues were taken as control.The expression of P gp and MRP1 were detected by immunohistochemical method.Results In gliomas the positive rate of P gp was 24 3%(9/37),but was 10%(1/10) in normal brain tissue.There was no statistics difference between them.The ratio of positive cells was less than 30% in positive specimens.The positive rate of MRP1 was 73%(27/37),whereas there was none in normal brain tissue.There was significance between them.P gp and MRP1 were seen in tumor blood vessel endothelium,the former being 24 3%(9/37),the latter being 35 1%(13/37).Conclusion Both P gp and MRP1 were expressed in gliomas,in which MRP1 was more than P gp.In tumor blood vessel endothelium the two proteins were abnormal,which indicated that blood brain barrier (BBB) might contribute to mutidrug resistance.
