摘要
目的 探讨致敏大鼠气道壁中成肌纤维细胞与Ⅲ型、Ⅴ型胶原的表达及成肌纤维细胞 (MF)在气道重建中的作用。方法 Wistar大鼠 3 2只 ,随机分为对照 4周组、8周组 ,致敏 4周组、8周组 ,每组 8只 ,并用卵白蛋白 (OVA)致敏并激发 ,制备致敏大鼠模型。对照组用生理盐水对照和激发。分别于激发 4周和 8周时取肺组织 ,用免疫组化法测定气道壁中Ⅲ型、Ⅴ型胶原和成肌纤维细胞、TGF β1的表达。结果 ①致敏 4周组和 8周组Ⅲ型胶原、Ⅴ型胶原和成肌纤维细胞的表达与对照 4周组和 8周组比较 ,差异均有显著性 (P均 <0 0 1) ;②致敏各组Ⅲ型、Ⅴ型胶原的表达与成肌纤维细胞的表达均呈显著正相关 (P均 <0 0 1) ;③致敏 8周组与 4周组比较 ,Ⅲ型、Ⅴ型胶原含量和成肌纤维细胞的表达显著增加 ,差异均有显著性 (P均 <0 0 1)。结论 哮喘气道重建过程中 ,有Ⅲ型、Ⅴ型胶原增生 ,成肌纤维细胞的过度表达 ,表明成肌纤维细胞促进了细胞外基质的形成 。
Objective To study the pathological features of airway remodelling in asthma in the rat and the function of myofibroblast in airway remodelling. Methods Thirty-two rats were randomly divided into 4 equal groups: 4-week normal control group, 8-week normal control group, 4-week asthmatic group and 8-week asthmatic group. Asthma models of rats were sensitized and challenged by ovalbumin(OVA), and control group by NS. The expression of collagen Ⅲ, Ⅴ and myofibroblast were detected with immunohistochemical ABC and imaging analysis methods. Results The collagen Ⅲ, collagen Ⅴ and myofibroblast stains were increased significantly both in 4-week and 8-week asthmatic groups compared with 4-week and 8-week control groups (P<0 01).The expression of collagens Ⅲ and Ⅴ were correlated with the expression of myofibroblast. Conclusion The increase of collagens Ⅲ and Ⅴ and the excessive expression of myofibroblast exist in airway remodelling, suggesting that myofibroblasts have an important role in airway remodelling.
