摘要
本实验采用了集落形成单位测定法观察抗微1号(KW-1)对γ-射线照射小鼠多能造血干细胞的保护作用。在照射前后各给KW-15天,能增加照射小鼠脾脏内源性造血灶的形成,同时,使脾脏重量和骨髓有核细胞数(NNBMC)较照射对照组明显增加,从而改善小鼠的整体造血功能;KW-1可显著加快照射小鼠多能造血干细胞(CFU-S)的增殖,使其恢复加快,4Gy照射后第14天,KW-1组CFU-S已恢复到正常水平的67.8%,而照射给水组则仅恢复到正常水平的40.7%;CFU-S剂量—存活曲线的结果表明,KW-1组与照射给水组之D_o(存活曲线上使存活细胞每减少63%所需要的照射剂量)和n(射线对靶子细胞的击中次数)无显著差异,说明KW-1对小鼠CFU-S的放射敏感性无明显影响。因此,KW-1防止照后效应,促进照射后CFU-S的增殖恢复是其抗辐射作用机制之一。
The protective effects of Kangwei-1 (KW-1) on multipotential hemopoietic stem cells in γ-Ray irradiated mice were observed with colony-forming unit assay. The administration of KW-1 for 5 days both before and after irradiation increased endogenous colony formation of the hemopoietic tissue on the surface of spleen, spleen weight and the number of nucleated bone marrow cells (NNBMC) in irradiated mice. It enhanced the recovery of the number of colony-forming unit of spleen (CFU-S) after irradiation by 4.0 Gy. The number of CFU-S recovered to 67.8 as much as of its physiological level on the 14th day after irradiation in KW-1 group, but only to 40.7 in the control group. The CFU-S dose-survival curve indicated that the differences of both 'D_0's (the radiation dose needed for reduction of survival cells on survival curve), and 'n's (the impact numbers of irradiation on the target cells) between the KW-1 group and the control group were insignificant, which denoted that KW-1 did not influence the radiosensitivity of CFU-S. The promoting effects of KW-1 on the hematopoiesis were one of the important mechanisms of its radioprotection.
