被动转移抗SIEA-Gp101血清对人工感染日本血吸虫鼠肺虫卵肉芽形成的影响

摘要 目的 了解SIEA Gp10 1对日本血吸虫虫卵肉芽肿形成的影响。方法 采用虫卵肉芽肿肺模型对SIEA Gp10 1的作用进行研究。将 36只昆明鼠随机分为 5组 ,各组被动转移的成分分别为 :抗SIEA Gp10 1,抗SIEA2 8kDa ,抗SIEA Gp10 1与抗SIEA2 8kDa混合血清 ,同时设正常兔血清组与生理盐水对照组。分别于第 1、3、5天尾静脉注射抗血清、正常兔血清或生理盐水 ,各组于第 2天注射日本血吸虫成熟虫卵 ,第 8天取鼠肺组织切片 ,观察虫卵肉芽肿变化并测量、比较虫卵肉芽肿大小。结果 正常兔血清组与生理盐水组相比 ,虫卵肉芽肿大小无统计学差别 (P >0 0 5 ) ;与正常兔血清组比较 ,实验组虫卵肉芽肿的大小分别下降了 18 0 8%、2 8 30 %和 6 44 % ,其中抗SIEA2 8kDa差别显著 (P<0 0 5 ) ,抗SIEA Gp10 1和混合血清组差别不显著 (P >0 0 5 )。肉芽肿内外细胞成分的观察显示 ,正常兔血清组与生理盐水组无明显的差别 ;而抗SIEA2 8kDa血清被转组 ,虫卵破坏加快 ,部分虫卵卵内仅见残渣和少量炎性细胞。抗SIEA Gp10 1与混合血清组部分虫卵坏死 ,有一定数量的炎性细胞浸润。结论 SIEA2 8kDa分子具有抗卵胚发育和抑制虫卵肉芽肿形成的作用。混合血清组较抗SIEA2 6 2 8kDa组虫卵破坏较轻 ,虫卵肉芽肿增大 ,细胞数量增多 。 Objective To study the effect of pulmonary egg granuloma formation of Schistosoma japonicum in mice transfused with anti-SIEA-Gp101 serum.Methods Thirty-six mice were randomly divided into five groups. A to E group were respectively treated with anti-SIEA-Gp101 serum, anti-SIEA28kDa serum, mixed sera, normal rabbit serum and normal saline. Anti-serum or normal saline were injected at day1,3,4,5.At day2,3000 mature eggs of S.japonicum were injected to each group of mice. Mice were killed at day8 day and their lungs were fixed in 10% formalin solution. The egg granulomas of the different group of mice were examined in the tissue sections stained with hematoxylin and eosin. The mean of granuloma diameter from each mouse was calculated and used to determine the average granuloma volume for each group. Comparisons of the results of different group were made using student's t-test.Results There was no significant difference in granuloma foration between the groups treated with normal rabbit serum and normal saline, In contract,the pulmonary egg granuloma size in the groups treated with anti-Gp101 serum,anti-SIEA28kDa serum and mixed sera showed reduction of 18.08%( P >0.05), 28.30% ( P <0 05)and 6 44%( P >0.05),respectively. In addition, the changes of egg and granuloma among the different groups were observed.Anti-SIEA-28kDa serum treatment facilitated the destruction of egg.Conclusion These results demonstrated that SIEA28kDa antigens could affect egg development and block the egg granuloma formation. Whether the increase in the intruding immune cells, the increase in sera of the granaloma and reduced in the egg destruction ability were due to the interference of anti-Gp101serum in the group treated with mix sera required further investigation.
