TGFβ_1在高氧暴露致肺纤维化早产大鼠肺组织不同细胞表达的定量研究

摘要 探讨转化生长因子β1(TGFβ1)与高氧暴露致肺纤维化发生、发展的关系及其在肺组织的细胞来源,应用免疫组化法结合图象分析处理系统定量研究TGFβ1在高氧暴露致肺纤维化早产大鼠肺组织不同细胞中的免疫反应。结果显示:TGFβ1可广泛分布于空气对照组早产大鼠肺组织,而在高氧组中表达明显增加。其中,肺泡巨噬细胞的表达在第7 d天达高峰(71.82±15.39),而第14 d支气管肺泡上皮、肺间质细胞的表达分布最广,阳性表达面积比分别为(46.47±9.65)和(54.39±12.35),此时肺组织羟脯氨酸含量明显增加[(2.08±O.12)mg.g肺-1]。结论:TGFβ1在高氧暴露致肺纤维化早产大鼠肺组织中具有重要作用,肺泡巨噬细胞TGFβ1的大量表达与肺纤维化的早期密切相关,而支气管肺泡上皮、肺间质细胞表达的进一步增强则在纤维化的持续进展中发挥了一定作用。 The relationship between TGFβ1 and pulmonary fibrosis and the resouce of TGFβ1 were studied in this investigation. We analyzed the immunoreaction of TGFβ1 of lung tissues of hyperoxia - induced pulmonary fibrosis of preterm rats with immunohistochemical techniques and image processing system. Result showed that the expression of TGFβ1 was widely distributed in bronchial epithelial cells throughout the air group preterm rats while it was increased in hyperoxia - induced rats. The expression in alveolar mac-rophages were maximumly elevated on day 7(71.82± 15.39, P <0.001). On day 14, the expression of TGFβ1 in bronchial epithelial cells and lung interstitium were maximurnly distributed, the ratio of positive area were46 . 47 ±9 . 65 , and54 .39± 12.35, respectively . When compared with the air group , P < 0.01, P< 0.001, respectively, When compared with 7 day group, P<0.01, 0.001, respectively. The content of oxyproline in hyperoxia injuried rats [(2.08 ±0.12) mg/g. lung]was increased compared with the air group (P< 0.001). We concluded that: TGFβ1, contributed significantly to hyperoxia- induced pulmonary fibrosis. Alveolar maerophages were concerned with the early stage while bronchial epithelium and lung interstitium might act on the progress of pulmonary fibrosis.
