中药复方连竹胶囊对糖尿病大鼠肾脏保护机制的研究

Protective Mechanism of Diabetic Nephropathy with Fufanglianzhu Capsule

目的 :建立糖尿病 (DM )大鼠模型 ,研究中药“复方连竹胶囊”对DM肾病的治疗和保护机制。方法 :将Wistar大鼠分为正常对照组、DM组和DM治疗组 ,DM大鼠模型由链脲佐菌素 (STZ)诱导。治疗 10周后 ,测定各组大鼠尿量、2 4h尿蛋白、尿素氮、血肌酐、糖化血红蛋白 (GHbAlc)、血清丙二醛 (MDA)和血浆NO含量 ,光镜下观察肾脏组织病理学改变。结果 :DM组的尿量、2 4h尿蛋白、尿素氮、血肌酐、GHbAlc和MDA均明显高于正常对照组 ,血浆NO含量低于正常对照组 (P <0 .0 1或P <0 .0 5 ) ,形态学观察可见DM组大鼠肾脏有弥漫性肾小球硬化表现 ,肾小球系膜区增宽、肾小管基底膜增厚等病理改变 ;DM治疗组的以上指标与正常对照组比较无明显差异 (P >0 .0 5 ) ,与DM组比较除NO外均显著降低 ,而血浆NO含量明显升高 (P <0 .0 5 ) ,肾脏病理改变较DM组有明显减轻。结论 :复方连竹胶囊对DM大鼠并发肾脏病变有较好的治疗作用 ,其机制可能与该方剂能抑制脂蛋白非酶基糖化、抑制脂质过氧化、促进NO生成。

Objective:To investigate the protective mechanism of Fufanglian zhu capsule on diabetic nephropathy.Methods:Wistar rats were divided into following groups:(1) control group;(2) DM group;(3)drug treated group.After diabetic rats were treated with Fufanglianzhu capsule for 10 weeks,we determined the concentration of GHbAlc,proteinuria,MDA,NO,BUA and serum creatinine and observed the renal pathological lesions.Results:Compared with the normal control group,there was a significant increase of levels of proteinuria,MDA,BUN,serum creatinine as well as GHbAlc,there was a decrease of the NO level ( P <0.01～0.05) in DM group .In drug treated group,GHbAlc,proteinuria,BUN,serum creatinine and MDA were reduced( P <0.05～0.01),the level of NO was increased( P <0.05).The renal pathological lesions in the drug treated group were less than those in DM group.Conclusion:Fufanglianzhu capsule have protective and cure effects on DN by inhibiting lipid peroxidation,reducing oxidant stess injury and increasing NO level.
