摘要
构建含HSV-TK基因的重组腺病毒AdTK,在两种肺癌细胞和鼠肺癌模型中对AdTK/GCV系统进行抑瘤实验研究。方法:采用同源重组法构建AdTK,用MTT法测定AdTK/GCV系统对H460、PG49人肺癌细胞的生长抑制率,建立T739鼠肺癌模型,观察AdTK/GCV系统对瘤体大小及鼠存活期的影响。结果:成功构建AdTK,AdTK转染H460、PG49细胞后对细胞抑制率随AdTK、GCV及药物作用时间的增加而增高,有显著性差异,小鼠体内实验中观察到AdTK/GCV系统对鼠肺癌有明显生长抑制作用,且明显延长了小鼠的存活期。结论:本文建立的AdTK/GCV系统在体内外对肺癌细胞均有明显抑制作用,为AdTK用于基因治疗奠定了基础。
Objective:In this reseach,the AdTK was constructed and identified and AdTK/GCV system cytotoxic effect on pulmonary carcinoma cells was assayed in vitro and in vivo.Methods:H460and PG49cell were infected with several concentration AdTK and GCV system and cell growth inhibition rate was evaluated by MTT method.AdTK was directly injected into established L795tumors-bearing T739mice for observing diffrence in tumor size and survival days of mice.Results:It is showed that the growth inhibition rate depended on AdTK dose,the concentration of GCV and the time of interaction in H460and PG49.We observed also that there was a significant difference in tumor size and survival days in mice between the therapy group and control group.Conclusion:AdTK/GCV system effectively suppressed pulmonary carcinoma cell growth in vitro and in vivo.It suggested that AdTK/GCV system may be very effective as a strategy for suicide gene therapy of pulmonacy carcinoma.
