体外免活性骨组织对同种骨髓基质细胞粘附特性影响的实验研究

摘要 目的 研究兔活性骨组织在共培养条件下对同种骨髓基质细胞粘附特性的影响。方法 分别应用生理盐水和三蒸水浸泡新鲜兔骨碎粒 5h后与同种骨髓基质细胞共培养作为实验组 ;未进行共培养的骨髓基质细胞作为对照组。应用体视学计数法观察接种后 0 .5、1、2、4、8h各时间点骨髓基质细胞贴壁情况 ,计量贴壁细胞数 ,计算出细胞贴壁率。结果 实验组的骨髓基质细胞贴壁率均显著高于对照组 (P <0 .0 1) ,但三蒸水浸泡的兔活性骨组织共培养的同种骨髓基质细胞的贴壁率最高。结论 活性骨组织在体外具有调节和促进同种骨髓基质细胞粘附特性的作用 ,用三蒸水浸泡的活性骨组织的作用最强。 Objective To investigate the effects of rabbit living bone compounds on adhesion characteristics of homogeneous bone marrow stromal cell in a co culture system. Methods In experimental group, rabbit fresh living bone compounds, immersed in 0.9% sodium chloride and distilled water for 5 hours, were used for co culture with homogeneous bone marrow stromal cells. The un cocultured rabbit bone marrow stromal cells served as the control. The attached cells were calculated by stereological method at hours of 0.5, 1, 2, 4 and 8 after seeding. Results The efficiency of attached cells was significantly higher in the experimental group than that in the control group( P <0.01); the efficiency of attached cells cocultured with the living bone compounds treated together with distilled water was the most significant. Conclusion Living bone compounds can have regulatory and accelerating effects on the adhesion characteristics of homogeneous bone marrow stromal cells in vitro . The effect of living bone compounds treated with distilled water on bone marrow stromal cells is the most significant. This experiment provides reference for the promotion of seeding cell adhesion on scaffold in the tissue engineering of bone.
