甲壳低聚糖对非肥胖性糖尿病小鼠胰岛细胞生长及胰岛素分泌量的影响 被引量:10

Effects of Chito-oligosaccharides on the Growth of Islets Cells and Insulin Secretion in Non-obese Diabetic Mice

摘要 目的探讨甲壳低聚糖对非肥胖性糖尿病 (NOD)小鼠胰岛 β 细胞生长增殖和释放胰岛素的影响。方法通过胰导管逆行灌注胶原酶静止消化NOD小鼠胰腺及不连续密度梯度Ficoll液纯化胰岛细胞 ,并用转板以及碘乙酸处理的方法除去其中的成纤维细胞 ,得到比较纯的胰岛细胞。以此为材料 ,将细胞分为干预组及对照组 ,干预组在细胞培养液中加入 3%甲壳低聚糖 ,对照组在细胞培养液中加入生理盐水。分别检测细胞增殖情况和急性胰岛细胞释放胰岛素实验。结果干预组细胞增殖优于对照组 (P <0 .0 5 ) ,而急性胰岛素释放则两组相比较差异无显著性。结论甲壳低聚糖能够促进胰岛 β ObjectivesTo explore the effects of chito oligosaccharides on proliferating activity and insulin releasing ability of pancreatic β islets cells in non obese diabetic(NOD) mice. MethodsThe isolation of pancreatic islets from NOD mice was carried out by stationary digestion after retrograde infusion of collagenase solution through pancreatic duct. The islets cells were purified by a Ficoll solution with discontinuous density gradient (25%,23%,20.5%,11%), then the fibroblasts were removed using iodoacetic acid method. The purified islet cells were divided into two groups: treating group and control group. 3% chito oligosaccharides were added to culture media of the former and 0.9% NaCl to those of the latter, respectively. The cellular proliferating activity was detected and acute experiment of insulin releasing from these islets cells was performed in both groups.ResultsThe cellular proliferating activity in the treating group was better than that in the controls (P<0.05). But there was no significant difference between the insulin releasing ability of two groups.Conclusion The chito oligosaccharides can promote proliferation of pancreatic β islets cells. [
