摘要
建立了一种非氰化物测定血红蛋白的新方法。该方法用季铵盐破坏红细胞膜 ,释放出血红蛋白 ,该血红蛋白与 EDTA结合 ,形成的血红蛋白衍生物在 53 5nm波长处有最大吸收 ,且在 4h内稳定 ,可用于血红蛋白的测定。初步探讨了反应机理。通过血样中血红蛋白的检测 ,并与氰化法所测结果对比 ,两者结果无显著性差异。
A new method has been established to measure hemoglobin by cyanide free reagent. It is based on that dodecyltnmethyl ammonium bromine breaks the membrane of red blood cells to give out hemoglobin, which combines with EDTA to form a new complex. The experimental result shows that when the amount of dodecyltnmethyl ammonium bromine is 3.0 mL and that of EDTA is 1.5 mL, the new complex can remain steady for 4 h, and the wavelength of absorption maximum is 535 nm. The reaction mechanism was preliminarily explored. The proposed method was applied to the measurement of hemoglobin in fresh blood, and the results obtained are coincided with that obtained by using cyanide reagent.
