IL-6、IL-10对骨髓基质细胞白血病抑制因子(LIF)的调节

The modulation of Interleukin-6 and Interleukin-10 on the expression of Leukemia inhibitory factor on the bone marrow stromal cells

目的研究白细胞介素-6(IL-6)、白细胞介素-10(IL-10)对骨髓基质细胞白血病抑制因子(LIF)表达的调节。方法用酶联免疫吸附试验(ELISA)测定IL-6和IL-10作用下的LIF的表达水平,用逆转录-聚合酶链反应(PT-PCR)测定IL-6和IL-10作用下的LIF转逆子水平。结果在IL-6作用下LIF水平明显降低(P<0.05);而IL-10对LIF水平没有明显影响。结论IL-6能下调骨髓基质细胞LIF的表达,而IL-10对骨髓基质细胞LIF的表达无明显调控作用。

Objective To study the effects of interleukin(IL)-6, IL-10 on the expression of Leukemia inhibitory factor (LIF) on the bone mairow stromal cells. Methods LIF protein level were measured by enzyme-Linked immunosorbent assays(ELISA). LIF mRNA were measured by reverse transcripton-polymerase chain reaction (PT-PRC) . Results LIF decreased significantly in groups of IL-6, LIF didn' t significantly change in groups of IL-10( P > 0.01) than that in the control group. Conclusion Being stimulated by IL-6, the expression of OF on the bone marrow stromal cells decreased. Being stimulated by IL-10, the expression of LIF on the bone marrow stromal cells didn't change.
