蛋白酪氨酸磷酸酶-1B基因多态性与2型糖尿病的相关性研究

摘要 目的 探讨蛋白酪氨酸磷酸酶 1B(proteintyrosinephosphastase 1B ,PTP 1B)基因 387位编码子Pro Leu多态性与 2型糖尿病 (T2DM )的关系。方法 采用多聚酶联反应 限制性片段长度多态性 (PCR RFLP)的方法对湖北地区 1 30例 2型糖尿病患者及 1 38例正常对照者PTP 1B基因 387位编码子酶切位点进行研究。结果  2型糖尿病患者和正常对照者PTP 1B基因均以PP基因型为主 ,其频率分别为 0 .94和 0 .96 ;P等位基因频率分别为 0 .97和 0 .98,差异无统计学意义 (P >0 .0 5)。 2型糖尿病患者中PTP 1B基因P387L变异与体重指数 (BMI)有关 ,但与空腹血糖、空腹胰岛素、总胆固醇、甘油三酯等临床变量不相关。结论 未发现PTP 1B基因 387位Pro Leu多态性与中国人 Obejective To investigate the 387 Pro Leu polymorphism in protein tyrosine phosphastase 1B (PTP 1B) and its relationship with the risk of type 2 diabetes mellitus(T2DM) in Chinese population. Methods The genotype and allele frequency of PTP 1B gene 387 Pro Leu polymorphism was assayed by polymerase chain reaction(PCR) restriction fragment length polymorphism (RFLP) between 130 T2DM and 138 normal controls. Results Both in T2DM group and control group ,the PP genotype of PTP 1B gene was the major one, and its frequencies were 0.94 and 0.96 respectively. There is no significant differences in the genotype and allele frequency of PTP 1B gene between T2DM group and control group(P> 0.05 ). The variation of PTP 1B gene in type 2 diabetes mellitus was related to body mass index, and it was not related to FBG,FINS,TC,TG or other clinical data. Conclusion The results suggested that the 387 Pro Leu polymorphism of the PTP 1B gene was not associated with type 2 diabetes mellitus in Chinese population.
