磷酰肌醇-3激酶途径在慢性髓细胞白血病细胞增殖和抗凋亡中的作用 被引量:1

摘要 目的 探讨磷酰肌醇 - 3激酶途径在K562和HL60细胞增殖中的作用。方法 用磷酰肌醇 - 3激酶(PI3K)特异抑制剂Wortmannin抑制PI3K活性 ,观察K562细胞和HL60细胞增殖能力的变化。结果 显示K562和HL60细胞在培养 2 4小时、 48小时、 72小时的增殖抑制率分别为 41 33 %、 57 46 %、 65 85 %和32 1 4 %、 1 7 1 4 %、 1 3 1 4 %。生长曲线显示Wortmannin可显著抑制K562细胞的增殖 ,而对HL60细胞增殖无明显影响。 Objective To explore the effect of phosphatidylinositol-3kin a se(PI3K)on the proliferation of CML cells.Method Wortmannin(i nhibitor of PI3K)was used,its action on K562 cells and HL60 cells was observed.Result The growth inhibition rates for K56 2 cells were 41.33 %,57.46%,65.85%respectively,and those for HL60 cells were 32.14%,17.14%,13.14% r espectively in 24 hours,48 hours,and 72 hours.Conclusion Wortm annin can inhibit the K562 cells proliferation through the PI3K pathway.
