摘要
目的 应用全反式维甲酸 (ATRA)作用前列腺癌DU - 14 5细胞、早幼粒白血病HL - 6 0细胞和乳腺腺癌MCF - 7细胞 ,观察维甲酸对多种肿瘤细胞生长的影响。方法 应用全反式维甲酸作用HL - 6 0细胞、DU -14 5细胞和MCF - 7细胞 ,分别以光学显微镜、电子显微镜观察细胞形态及超微结构的改变 ,并用流式细胞仪检测细胞周期的改变。结果 全反式维甲酸作用前列腺癌DU - 14 5细胞、早幼粒白血病HL - 6 0细胞和乳腺腺癌MCF - 7细胞后 ,导致肿瘤细胞生长减缓 ,肿瘤细胞超微结构出现细胞核固缩、核碎裂、染色质凝集于核膜周边等细胞凋亡的典型形态学改变 ,流式细胞仪检测肿瘤细胞出现亚二倍体“凋亡小峰”。结论 全反式维甲酸可以诱导多种肿瘤细胞产生凋亡 。
objective To treat prostatic carcinoma cell line DU-145?breast cancer cell line MCF-7 and promyeelocytic cell line HL-60 by all trans retinoic acid(ATRA), to observe the change of tumor cell's growth. Methods DU-145 cells?HL-60 cells and MCF-7 cells were treated with ATRA(10-5mol/L)for 36h and 72h, observed changes of the cell's shape and ultrastructure by optical and electron microscope. Analyzed the effect of retinoic acid on the cycle of tumor cells by flow cytometry FCM. Results After treated with ATRA, The results showed that all kinds of carcinoma cells DU-145 grow slowly, some of the carcinoma cell's ultrastructure appeared early apoptotic changes, including cell shrinkage, nuclear condensation. After 72h treated with ATRA, some apoptotic cells were detected,and a typical subdiploid peak were observed by flow cytometry. Conclusions Retinoic acid can induce multiple carcinoma cells to apoptosis. The apoptotic process has a time -depending character, and also depand on the concentration of retinoic acid.
