孕鼠子宫平滑肌与左心室心肌细胞L型电压依赖型钙通道基因表达的变化

摘要 目的:探讨孕鼠子宫平滑肌与左心室心肌细胞L型电压依赖型钙通道基因(VDCC-L)的功能性亚单位α1、β2mRNA表达与分娩的关系。方法:应用半定量逆转录聚合酶链反应(RT-PCR)分别测定未孕鼠、晚孕鼠、产时鼠、产后鼠子宫平滑肌与左心室心肌VDCC-L α1、β2 mRNA相对表达量的变化。结果:4组子宫平滑肌细胞VDCC-Lα1、β2 mRNA表达量,α1依次为0.2911±0.1777、0.4376±0.1342、0.8003±0.1659和0.3942±0.1503,β2依次为0.3045±0.1542、0.3168±0.1459、0.6469±0.1304和0.3202 ±0.1239;α1、β2mRNA在分娩前后的表达规律相似,晚孕鼠较未孕鼠表达增加,其差异均无统计学意义(P>0.05),产时表达急剧升高,与未孕鼠、晚孕鼠相比,差异均具有显著性意义(P<0.05),产后则急剧减少,与产时相比,差异均有显著性(P<0.05)。心肌VDCC-L在4组的表达量,α1依次为0.3020 ±0.1140、0.5033±0.1641、0.6625±0.1801和0.6065±0.1482,β2依次为0.3288±0.1140、0.5250±0.1711、0.6702±0.1405和0.5748±0.1523;孕期有所增加,晚孕鼠与未孕鼠相比差异无统计学意义(P>0.05);产时表达进一步增加,与未孕鼠相比,差异具有显著性意义(P<0.05),与晚孕鼠相比,差异无显著性(P>0.05);产后24h仍维持较高水平,与产时相比无显著性差异(P>0.05)? Objective:To study the relationship between expressions of VDCC-L α1、β2 mR-NA in the pregnancy rat uterine smooth muscle and left ventricular myocardial cells and parturition. Methods: A reverse transcription-polymerase chain reaction(RT-PCR)method was utilized to examine and quantitate VDCC-L α1、β2 mRNA expressions in the uterine myometrum and left ventricular. Forty rats were divided into non-pregnancy x late pregnancy x laboring and after laboring four groups, each with 10 cases. Results: (1)In the uterine myometrium, the levels of VDCC-L α1 mRNA expression in the non - pregnancy , late pregnancy x laboring and after laboring group were 0.2911 + 0.1777、0.4376 ± 0.1342、0. 8003 ± 0.1659 and 0.3942 ± 0. 1503, respectively. The levels of VDCC-Lβ2 mRNA expression in the above four groups were 0. 3045 ± 0.1542、0. 3168 ± 0.1459、0.6469 ±0.1304 and 0.3202 ± 0.1239,respectively;There was no remarkable difference between the non-pregnancy group and late pregnancy group ( P > 0.05) . The Levels in laboring group were significantly higher than that in late pregnancy group or non-pregnancy group(P < 0.05),but in af- ter laboring group decreased rapidly,remarkable difference was found between laboring and after la— boring group(P<0.05).(2)In the leg ventrieular myocardial cells,expressions of VDCC—L al in the non—pregnancy、latepregnancy、laboringandafterlaboringgroups wereO.3020±0.1 1 40、 0.5033±0.1641、0.6625±0.1801、0.6065±0.1482,respectively;lhe levels of VDCC.L&mRNA expression wereO.3288±0.1140、0.5250 4-0.1711、0.67024-0.1405、0.57484-0.1523,respectively. Although both the levels of VDCC—L田and VDCC—Lf32 mRNA expression in late pm4pamaey group were a little higher than that in the non-pregnancy group,the statistic difference was not noted(P>O.05), the significant difference lied in laboring group to non-pregnancy group(P<0.05);the higher expres sion in late pregnancy、laboring and after labo,~g groups,no difference WaS showed remarkably (P>0.05).Conclusiom:The high coexpmssions ofVDCC—L al、&mRNA in the uterine myometnan at laboring rat show a close correlation with the pama4tion by joining many steps.Levels in left ven— trieular myocardial ceils at before and after laboring reveal their relationships to the heart function’s changing.
