摘要
目的:建立较为全面反映肝星状细胞(hepatic stellate cell,HSC)活化效应的细胞模型。方法:应用脂多糖(lipopolysccharide,LPS)刺激体外培养的大鼠HSC。噻唑蓝(MTT)法测定HSC增殖率。免疫组化法显示HSC表达其Ⅰ、Ⅲ型胶原的表达水平,并进行图象分析。结果:LPS明显刺激HSC的增殖并提高Ⅰ、Ⅲ型胶原表达水平,与对照组比较差异有非常显著性意义(P<0.01)。结论:LPS诱导HSC活化模型能够相对全面地反映HSC活化的效应。
Objective: Development of cellular model 1 of HSC activation. Methods: Isolated rat HSC cultured in vitro was stimulated with lipopolysaccharide (LPS). Proliferation of rat HSC was detected with MTT technique. Expression levels of collagen I and collagen Ⅲ in rat HSC were showed with immunochemistry technique and analyzed with image analysis system. Results: LPS significantly promoted proliferation elevated expression levels of collagen I and collagen M in rat HSC. Conclusion: Model of rat HSC activation stimulated with LPS can reflect more intact HSC activation effects.
