次甲基兰水溶液的最大吸收波长为663.5nm。肝素与次甲基兰水溶液反应后,其吸光度A的降低与肝素浓度(效价)成正比。溶液颜色稳定,采用比较法测定。肝素浓度在0.01-).05mg/25ml范围内服从朗伯比尔定律。平均回收率99.73%。变异系数0.53%。该法可用于肝素钠注射液的效价测定。
The maximum absorption wavelength of methylene blue aqueous solution is 663.5nm. Decrease in absorbance was in proportion to heparin concentration (potency) after the interaction between heparin and methylene blue. The solution colour was stable. The excellent linearity was obtained at the heparin concentration ranging from 0.01 to 0.05mg/25ml. The average recovery was 99.73%. The variation coefficiet was 0.53%.The method can be applied to determine the potency of heparin sodium injection.
