摘要
目的:考察不同制样方法对透射电镜观测微粒给药制剂的影响。 方法:制备含有乳剂的琼脂小胶囊 ,分别采用醋酸铀直接负染法、锇酸固定法及戊二醛、锇酸双重固定法 ,观测去氢骆驼蓬静脉注射用乳剂的微观形态 ;分别采用负染法和冷冻蚀刻技术制备样品 ,观测替加氟脂质体的微观形态。 结果 :先制备琼脂小胶囊 ,再用戊二醛、锇酸双重固定后染色 ,可得亚微型静脉乳的清晰图像 ;采用冷冻蚀刻技术制备样品 ,可观测脂质体微粒的三维结构构象 ,成像清晰。 结论:应用透射电镜观测微粒给药制剂时 ,可根据观测对象特点 。
Objective: To study the influence of different sample making method in observation of particles drug medication agent with transmission electron microscope(TEM) method. Methods: With making agar microcapsules containing emulsion at first, negative staining technique using uranyl acetate directly, technique using osmiue tetroxide fixed, and technique using glutaraldehyde and osmiue tetroxide fixed were employed to observe microcosmic form of harmine intravenous emulsion; negative staining technique and freeze etching technique were adopted to observe microcosmic form of tegafur liposome. Results: Darity figure of subminiature intravenous emulsion was obtained with making agar microcapsule firstly and staining after double fixing with glutaraldehyde and osmiuete tetroxide, then three dimensional structure conformation was abstained with making sample using freeze etching method. Conclusion: When using transmission electron microscope to observe particles medication agents, we can choose sample making method freely according to characteristics of objectives abserved.
