卵巢癌细胞中小分子G蛋白Rho的表达及其意义 被引量:3

摘要 目的 研究小分子G蛋白Rho家族中RhoA、RhoC及其效应分子ROCK -1在不同卵巢癌细胞中的表达水平 ,探讨它们与卵巢癌转移的相关性。方法 RT -PCR检测RhoA、RhoC及ROCK -1在卵巢癌细胞SW62 6、Skov-3、A2 780和Caov-3中mRNA的表达水平 ;WesternBlot检测RhoA和ROCK -1蛋白的表达情况 ;Boyden小室体外侵袭试验检测四株卵巢癌细胞的体外侵袭能力。结果 RhoA、RhoC和ROCK -1在不同的卵巢癌细胞中均有表达 ,但仅RhoC的表达水平与癌细胞侵袭力能力呈正相关 (r =0 .95 ,P <0 .0 1 )。此外RhoA在转录水平和蛋白水平并不呈同步变化。四株卵巢细胞的体外侵袭能力 ,其中SW62 6最强 ,Caov-3最弱 ,Skov -3和A2 780居中 ;结论 RhoC的表达与卵巢癌细胞的体外侵袭能力相关 ,可能作为一个癌基因在卵巢癌中起作用 。 Objective To investigate the role of RhoA、 Rh oC and their effector ROCK-1 of small molecular G protein Rho in motility and in vasion in ovarian tumor cells. Methods The expression of RhoA、RhoC and ROCK-1 in different ov arian tumor cell lines SW626、Skov-3、A2780 and Caov-3 were analysed by using se miquantitative RT-PCR and Western blot.Boyden chamber in vitro invasion assay w as used to evaluate these cells' invasive capability. Results RhoA、RhoC and ROCK-1 w ere all expressed in four ovarian tumor cell lines,but RhoC is the only one tha t was directly correlated to these cells' invasive capacity.Expression of RhoA in transcription level didn't concide with that in translation level. Conclusion There was a strong direct correlation between the l evel of RhoC mRNA expression and the invasive capability of all four ovarian can cer cells in vitro experiments,so it may serve as an oncogene in ovarian cancer and a new target in the diagnosis of early ovarian tumor.
