摘要 目的建立一种操作简单、快速、灵敏度高、重现性较好的测定低浓度蛋白质含量的方法。方法研究Folin 酚试剂法对低浓度蛋白质含量测定的可行性 ,并将其与考马斯亮蓝染色法在一定浓度范围 (<5 0 μg ml)内进行方法学比较。结果Folin 酚试剂法对牛血清白蛋白在 5~ 5 0 μg ml浓度范围内测得吸收度值均较小 (<0 .1 5 ) ,但示数稳定、线性关系好、精密度高、准确度好 ,并且以牛血清白蛋白为对照蛋白测定蛋白质供试品测定结果较考马斯亮蓝染色法高。结论Folin 酚试剂法适合于低浓度 (<5 0 μg ml)蛋白质溶液中蛋白质含量测定 。 Purpose To establish a simple, rapid, sensitive and accurate method to determine the content of low concentration protein solution.MethodsThe feasibility of Folin phenol protein quantification method for low concentration protein was investigated and compared with Coomassie Brilliant Blue (CBB) binding method.ResultsThe absorbence of bovine serum albumin (BSA) within the concentration range of 5 to 50 μg/ml in Folin phenol protein quantification method was small, but the method had good reappearance, linearity and accuracy. The determination result of Folin phenol method was much higher than that of CBB binding method when BSA was used as reference protein.ConclusionFolin phenol method can be used to determine the content of low concentration (<50 μg/ml)protein and is better than CBB binding method.
