大鼠大脑中动脉缺血再灌注损伤细胞凋亡及Bcl-2分布情况的观察 被引量:1

摘要 目的:观察大鼠大脑中动脉缺血再灌注后脑组织的病理学变化、细胞凋亡及Bcl—2表达情况。方法:利用腔内线栓法制备大鼠大脑中动脉闭塞模型,采用HE染色、免疫组化方法及TUNEL法观察缺血侧大脑半球神经细胞坏死、Bcl—2分布及细胞凋亡分布情况。结果:(1)坏死的神经细胞出现在缺血侧大脑半球的基底节区、额顶叶皮质及海马区。(2)凋亡的神经细胞大量出现在坏死灶边缘区,坏死灶内部也有少量凋亡细胞。(3)Bcl—2阳性神经细胞分布于远离坏死灶的周边区域,其细胞形态正常,分布范围超出HE染色异常区。结论:(1)细胞凋亡是急性脑缺血损伤细胞死亡的重要形式。(2)Bcl—2阳性表达区域可能是半暗带在光镜下可以定位到的部分区域。 Objective: To study the apoptosis and the positive expression of Bcl-2 of neurons in focal cerebral ischemia-reperfusion injury models of Wistar rats . Methods: The models were established by occluding middle cerebral artery with intraluminal sutures . HE staining , immunohistochemistry and TUNEL methods were used to observe the necrosis , apoptosis and expression of Bcl- 2 of neurons in ischemic cerebral tissue. Results: The necrotic neurons appeared in the ischemic basal ganglion area , frontoparietal cortex and hippocampus . A large number of apoptotic neurons were located in the margin of necrotic area and a few apoptotic neurons also could be found in the inner necrotic area . Microscopically , the neurons with positive expression of Bcl- 2 failed to be found in changes in cell morphology . Conclusion: Apoptosis is an important form of neurons death and the area with positive expression of Bcl-2 perhaps is a part of penumbral tissue which can be observed by light microscope .
