高压氧加抗氧化剂能减轻大鼠肝损伤、迟滞肝纤维化发展 被引量:1

摘要 目的 探讨高压氧加抗氧化剂对实验性大鼠肝脏损伤及胶原产生的影响。方法 用四氯化碳诱导大鼠实验性肝纤维化模型 ,将 2 4只雄性Wistar大鼠随机分为正常对照组 (C) ,模型组 (M) ,高压氧加抗氧化剂组 (HFr) ,秋水仙碱组 (PC)各 6只 ,8周后处死 ,观察肝组织病理变化 ,免疫组织化学方法检测I、Ⅳ型胶原在肝组织中变化及分布情况 ,并进行半定量分析比较。结果 与模型组比较 ,高压氧加抗氧化剂组大鼠肝重指数 (肝重 /体重 )降低明显 (P <0 .0 1 ) ;在实验各组中血清转氨酶活性最低 ;肝组织I、Ⅳ型胶原蛋白表达明显低于模型组 (P <0 .0 5 )。结论 高压氧加抗氧化剂可显著减轻实验性大鼠肝损伤以及I、Ⅳ型胶原的蛋白表达 。 Objective To investigate the effects of Hyperbaric oxygen and anti-free agency on the treatment of the liver lesion and the expression of type Ⅰ、Ⅳ collagen proteins in experimental hepatic fibrosis in rats.Methods Twenty-four male Wistar rats were randomly divided into four groups,including normal control group,hepatic fibrosis model group,Hyperbaric oxygen and anti-free agency group and cholchicine group.Experimental liver fibrosis was induced in rats by subcutaneous injection of carbon tetrachloride(CC14)oily solution for a period of 8 weeks.Using Hematoxylin-eosin staining technique,we observed the pathomorphological change of hepatic tissue under light microscope,compared the degrees of proliferation of collagenous fibers,and assessed the expression of collagen Ⅰ、collagenⅣ by immunohistochemical staining.Semi-quantitative scores were used to evaluate the degree of fibrosis.Results The liver weight index in Hyperbaric oxygen and anti-free agency group was lower than that in mode group.The levels of ALT、AST in serum of Hyperbaric oxygen and anti-free agency group were lowest.And the expression of collagen Ⅰ、collagen Ⅳ was significantly decreased in Hyperbaric oxygen and anti-free agency group than model group(p<0.01,p<0.05).Conclusion The results are suggested that Hyperbaric oxygen and anti-free agency can reduce liver lesion and theexpression of hepatic type Ⅰ、Ⅳ collagen proteins,and play an important role in against hepatic fibrogenesis.
