HPLC-MS/MS法快速测定阿胶中4种主要氨基酸

Rapid Determination of 4 Main Amino Acids in Colla Corii Asini by Liquid Chromatography Tandem Mass Spectrometry

目的:建立阿胶中羟脯氨酸、甘氨酸、丙氨酸、脯氨酸4种氨基酸的液质联用快速测定方法。方法:样品以6mol·L^(-1)盐酸于150℃水解1小时,旋转蒸干后用0.01mol·L^(-1)盐酸溶解,过滤后直接采用液质联用色谱法,于电喷雾源(ESI)正离子模式下使用选择性反应离子扫描(SRM)进行定量。结果:该方法抗干扰能力强,检测快速,四种氨基酸的加样回收率(n=6)为97.2%~103.7%,相关系数0.9973~0.9999。结论:此方法操作简便、快速准确,具有良好的重复性和稳定性,可用于阿胶中羟脯氨酸、甘氨酸、丙氨酸、脯氨酸含量的快速检测。

To establish a rapid and simple method for determining 4 amino acids(hydroxyproline,glycine,alanine,proline)in colla corii asini by HPLC-MS/MS.Method;The sample was hydrolyzed for 1 hour at 150℃,dried by rotation evaporation,and dissolved by 0.01 mol · L^(-1) sodium acetate.After filtration,HPLC MS/MS method was adopted.The reactive ion scanning was selected in positive ion mode by electrospray ionization(ESI) to quantitatively analyze amino acids.Result;The rapid method had strong anti-jamming ability.The recoveries were between 97.2%and103.1%,and the calibration curves of the amino acids were excellent with correlation coefficients between 0.9973 and0.9999.Conclusion;The method established in this paper is accurate with high repeatability,which is helpful for the determination of hydroxyproline,glycine,alanine,proline,4 amino acids in colla corii asini.
