摘要 本文提供一种制备肝剖面扫描电镜标本的改进方法并报告应用此法研究大鼠肝脏的结果。主要的实验改进是在制备肝剖面时引入一个新的步骤:先用含EDTA的细胞分离液短时间灌流肝脏后,再进行固定和其它处理。本文方法有如下的优点:1)在肝剖面上肝细胞基本无破损,2)能完好地原位暴露肝细胞的血窦面,3)胶原纤维结构保存比较好。它特别适合于观察肝组各种成分之间的相互位置关系。我们直接观察到成纤维细胞和胶原纤维在肝细胞表面的分布情况,这可能是文献中的首次报导。 We present here an improved method for scanning electron microscopy of liver dissec-tion surface specimen and report the result of its application to rat liver. The major experimental im-provement was introducing a new step in preparating dissection surface by a short-time perfusion of liverwith an EDTA-containing fluid. The experimental procedures were discribed and the result of SEM study was reported (chiefly inphotos). The stated method especially suit to study topographic relationship between liver tissue elementsand more attention had paid to the distribution of fibroblasts and collagen fibers on hepatocyte surface. Itcould be a new progress on this respect.
