大麦耐湿性的SSR标记与初步定位研究

摘要 湿害是严重影响大麦生产的重要因素之一,目前关于湿害基因的分子标记研究还较少。以耐湿大麦品种Stiring和不耐湿的大麦品种京裸11为亲本构建F2群体,对亲本及F2单株进行耐湿性鉴定,结果表明,倒二叶的叶绿素含量可作为耐湿性的鉴定指标。选择极端耐湿和不耐湿的F2单株组成耐湿、感湿两个池,利用73对引物对其进行PCR扩增检测多态性,结果发现引物WMC1E8在两亲本和两池间均检测出多态性。利用群体进行分析,表明WMC1E8标记位点与耐湿性相关极显著,可解释耐湿性表型变异的29.91%,初步可以认定该位点位于大麦第一染色体上。 Water-logging affects seriously the production of barley. However, researches on genetic mapping of water-logging tolerance in barley were rarely reported. 136 F_2 population were developed from a cross between Stiring (water-logging tolerance) and Jingluo11 (susceptible to water-logging) in order to identify DNA markers for genes conferring water-logging tolerance. The correlation between content of chlorophyll in the second top leaf and water-logging tolerance was very significant. It could be used as a phenotypic marker for water-logging tolerance in barley. The F_2 population displayed a two-peak distribution with different content of chlorophyll. Two DNA bulks were constructed based on content of chlorophyll in F_2 population. 73 SSR primers were used to screen polymorphism for parents and two DNA bulks by PCR amplification. The marker WMC1E8 showed polymorphism between two parents and between tolerant and susceptible bulks. This locus showed significant correlation to water-logging tolerance of the population, explaining 29.91% of total phenotypic variation.
