摘要
为获得稳定遗传的高直链淀粉含量转基因株系,并为下一步转基因小麦的转基因安全评价"环境释放试验"做准备,以本研究室前期获得的抑制淀粉分支酶基因sbeⅡa表达的转基因小麦株系4-4-1、4-4-2、4-4-3、4-4-19的T4代植株为试材,在北京市进行了一年的"中间试验",结果表明,在田间栽培条件下,转基因小麦植株个体生长发育正常,生长发育各个阶段所需的天数与对照基本一致,生长特性没有发生变化,但转基因小麦在株高、分蘖数、小穗数、不育小穗数等性状上与对照存在差异。4个转基因株系的千粒重均显著低于对照,4-4-1、4-4-3、4-4-19的株高均显著低于对照,4-4-2和4-4-19的小穗数显著多于对照。此外,灌浆期籽粒内源sbeⅡa基因的表达、SBE活性和品质性状检测结果表明,外源RNAi构件能够稳定遗传,功能得到正确表达。
The T4 transgenic wheat lines 4-4-1,4-4-2,4-4-3 and 4-4-19 with sbeⅡagene knocked down were evaluated under field conditions in 2010.The results showed that the plant growth and development of all transgenic lines were normal under the test conditions.But their morphological and major agronomic traits were different from the wild receptor plants,such as plant height,number of tillers,spikelets,spikelets infertility etc.The grain weight of the four transgenic lines was lower than the wild type.The plant height of the transgenic wheat lines 4-4-1,4-4-3 and 4-4-19 was lower than the wild type.The transgenic wheat lines 4-4-2 and 4-4-19 had more spikelets.Analysis on the expression of endogenous sbeⅡ agene,SBE activity and quality traits showed that the target gene was stably inherited.
