摘要
目的:研究正常皮肤和瘢痕组织中乏氧诱导因子-1α(HIF-1α)和促炎细胞因子的水平及二者相关性。方法:选取2015年3月-2017年3月在笔者医院切除的四肢瘢痕疙瘩组织标本为瘢痕疙瘩组;增生性瘢痕组织标本为增生性瘢痕组;普通瘢痕组织标本为普通瘢痕组,同期因四肢外伤切除的正常皮肤组织作为正常组。研究采用HE染色观察炎性细胞浸润情况,RT-qPCR检测病理性瘢痕组织和正常皮肤组织中HIF-1α及促炎细胞因子的蛋白表达水平。结果:瘢痕疙瘩观察组及增生性瘢痕组浸润水平明显高于普通瘢痕组及正常皮肤组(P<0.05),普通瘢痕组与正常皮肤组浸润水平无明显差异(P<0.05),瘢痕疙瘩组浸润水平明显高于增生性瘢痕组(P<0.05)。瘢痕疙瘩组及增生性瘢痕组中HIF-1α、IL-6、IL-8、hs-CRP的mRNA表达水平显著高于正常皮肤对照组(P<0.05),瘢痕疙瘩组HIF-1α、IL-6、IL-8、hs-CRP的mRNA表达量显著高于增生性瘢痕组(P<0.05)。HIF-1α的表达与炎性细胞因子IL-6、IL-8、hs-CRP呈正相关(P<0.05)。结论:病理性瘢痕组织中HIF-1α和促炎细胞因子水平显著高于正常皮肤,且HIF-1α和促炎细胞因子的水平呈正相关性。
Objective To study the levels of hypoxia-inducible factor-1α(HIF-1α) and pro-inflammatory cytokines in normal skin and scar tissue and their correlation. Methods The keloid tissue of the extremities excised from the author’s hospital from March 2015 to March 2017 was selected as the keloid group;the hypertrophic scar tissue specimen was the hyperplastic scar group;the common scar tissue specimen was the common non-pathological scar group. The normal skin tissue removed by extremity trauma was used as the normal group. The inflammatory cell infiltration was observed by HE staining. The protein expression levels of HIF-1α and proinflammatory cytokines were detected by RT-qPCR in pathological scar tissue and normal skin tissue. Results The infiltration level of the keloid group and the hypertrophic scar group was significantly higher than that of the normal scar group and the normal skin group.(P<0.05). There was no significant difference in the infiltration level between the normal scar group and the normal skin group(P<0.05). The level of infiltration in the observation group was significantly higher than that in the hypertrophic scar observation group(P<0.05). The mRNA expression levels of HIF-1α,IL-6,IL-8 and hs-CRP in the keloid group and the hypertrophic scar group were significantly higher than the normal skin control group(P<0.05), the mRNA expression levels of HIF-1α,IL-6,IL-8 and hs-CRP in the keloid group were significantly higher than those in the hypertrophic scar group(P<0.05);the expression of HIF-1α was positively correlated with proinflammatory cytokines IL-6, IL-8 and hs-CRP(P<0.05). Conclusion The levels of HIF-1α and proinflammatorycytokines in pathological scars were significantly higher than those in normal skin, and the levels of HIF-1 α and proinflammatory cytokines were positively correlated.
